Qianyang Jiedu Tongluo Drink On The Protective Effect And Mechanism Of Cardiac And Renal Target Organ Of Shr

Objective: The effects of different doses qianyang Effects of JieDuTongLuo drink in spontaneously hypertensive rats (SHR) blood pressure,left ventricular mass index (LVWI), left ventricular morphologic, left ventricular ultrastructure of myocardial connective tissue growth factor (CTGF) mRNA expression and kidney AT1, AT2mRNA expression to explore the protective effect of hypertensive target organ and TCM pathogenesis.Methods: Male 12-week-old spontaneously hypertensive rats, weighing 220±30g between the model were randomly divided into blank control group (SHR-C), enalapril (SHR-Y), traditional Chinese medicine high dose group (SHR -Z1), traditional Chinese medicine middle dose group (SHR-Z2), traditional Chinese medicine low dose group (SHR-Z3). 10 rats in each group. Harvested after 4 weeks of administration measured before and after administration of rat tail artery systolic pressure, left ventricular mass index and cardiac observation of morphological changes were detected with the meristem of myocardial connective tissue growth factor (CTGF) mRNA expression and renal tissue AT1, AT2mRNA expression..Results:1. As the weeks of increases, the blank control group, traditional Chinese medicine low dose group of the SBP gradually increased SHR. After 4 weeks of administration of Chinese medicine high,middle dose group, enalapril group compared with blank control group, SBP, Chinese medicine low dose group was significantly lower (p <0.05); medicine high-dose group, middle dose group, enalapril group LVWI lower than those in the control group (p <0.01), low-dose of Chinese medicine, compared with the control group, no significant difference (p> 0.05).2. Light microscopy showed: high-dose group and the enalapril group the majority of normal cardiac fibers, no difference between the two groups than in the low-dose group better. Blank control group that most myocardial fibers thicker, and some visible muscle fiber fracture, disordered, some nuclei hypertrophy. TEM observation showed: blank control group, and traditional Chinese medicine low dose group were irregularly arranged filaments, some of myofilament dissolution, fracture, muscle sections with light and dark is unclear, mild separation of intercalated disc, part of the increase in mitochondria, cristae broken, irregular-shaped nucleus . Medicine of high, medium-dose group, enalapril significantly reduced the above-mentioned diseases. 3. Compared with the group, blank SHR rat myocardial tissue expression of CTGF mRNA increased (P <0.01), where the positive control group and Chinese medicine high, middle dose group CTGF mRNA expression was significantly decreased (P <0.01); medicine low-dose group than CTGF mRNA expression in SHR rat groups (P> 0.05).4. Positive drug enalapril group kidney AT1 mRNA expression was significantly increased, while taking Qian Yang Effects of JieDuTongLuo drink later, with the dose increased renal AT1 mRNA expression in tissues is significantly increased, showing clearly the amount of effect relationship. SHR kidney tissue AT2mRNA weakly expressed in positive enalapril group can significantly increase the AT2 mRNA expression, while taking Qian Yang Effects of JieDuTongLuo drink later, with the dose increase in AT2 mRNA in kidney expression also increased significantly, showing a clear dose-response relationship, the trend with the Qian Yang Effects of JieDuTongLuo drink on the effects of AT1 mRNA. Fig2 with the results compared with the SHR, Qian Yang Effects of JieDuTongLuo drink can significantly increase the ratio of AT2/AT1.Conclusion:1. Qian Yang Effects of JieDuTongLuo SHR drinking can significantly reduce the SBP, to reduce LVWI. And showed the dose-effect relationship.2. Hypertensive target organ protection and TCM pathogenesis may be related to drug-related harm into the network. Qian Yang Effects of JieDuTongLuo drink can slow the SHR target organ damage, to prevent myocardial and kidney damage, reduce the myocardial connective tissue growth factor (CTGF) mRNA expression in kidney tissue and increase AT1, AT2mRNA expression may be qianyang Effects of JieDuTongLuo drink to improve the Hypertensive target organ damage mechanisms.
