Acupuncture Treatment Of Chronic Prostatitis Hyperplasia And Its Mechanism Exploring

Objective:A Pathological Model of Rats with chronic and proliferative prostatitis was established to study the acupuncture effect to serum TNF-a and IL-6on the modelMethod:(1) Model of chronic and proliferative prostatitis:Selected male SD rats as model animal, after been anesthetized with10%hydration chlorine aldehyde (300mg/kg), removed the hair with4%sodium sulfide. Then open peritoneal cavity by sterile working and inject hemorrhoid injection0.2ml to left and right lobe of prostate, while injecting equal-volume physiological saline into sham operated control group rats, and suture abdominal wall muscles and skin. Ingestation freely after12h for operation, and sterilized wound to prevent infection.(2) Animal group: The Rats were randomly divided into model group、drug group、acupuncture group and drug-acupuncture group when the the model were determined succeed. Meanwhile, the sham operated rats as health group.(3) The acupoints choice:Chinese medicine dialectical Chronic proliferative prostatitis for spleen and kidney deficiency, and the hot and humid stasis sluggish for standard. So selected the guanyuan, taichong、sanyin、sanyinjiao、piyu and shenyu five points to acupuncture treatment.(4) Physic liquor preparation:Choose Qianlietong tablet as drug treatment which preparated into0.1g/mL solution according to people and SD rats drug conversion.(5) Test treatment:Acupuncture group:When the rats were been fixed on the stage, use the meridians detectors get the needle points and acupuncture the guanyuan、shenyu、 sanyinjiao、piyu、taichong piont with the0.28mm×25mm HaoZhen in sequence, one day once,10days for one period of treatment. Drug-acupuncture group: Treatment with acupuncture group the same, and each rats filled2ml Qianlietong solution every day at the same time; Drug group:each rats filled2ml Qianlietong solution every day; The health group and model group just been fixed on the stage having nothing processing.(6) Observe the pathological changing:put the rats to death after take the blood, and gather the prostate maded into tissue slice when each group haved the first, second and third period of treatment, each group of5rats only. (7) Determined the serum TNF-a and IL-6:Separate the serum and determin the TNF alpha and IL-6when each group haved the first, second and third period of treatment, each group of5rats only.Results:(1) Model group:The prostate gland bubble structure chaos, part of the lumens deformation or disappear, visible glandular epithelium cells necrosis and fall off, interstitial edema and there had lots of inflammatory cells infiltrating diffuse, interstitial fibre hyperplasia obviousily, more fibroblasts, vascular dilatation and congestion by light microscopy. The drug group、acupuncture group and drug-acupuncture group:Prostate tissue structure tend to be normal, no obvious edema and hyperplasia, accidentally see inflammatory cells after three course of treatment. The drug-acupuncture group’s prostate tissue structure were more normal than drug group and acupuncture group’, cupuncture group’s prostate tissue structure were more normal than Drug group.(2) the model group’s TNF-a and IL-6levels increased significantly compared with health group, having a significant difference (P<0.01), the drug group、acupuncture group and drug-acupuncture group’TNF-a and IL-6levels reduced significantly compared with health modle group, having a significant difference (P<0.01).Conclusions:Acupuncture can promote prostate tissue returned to normal and reduce the serum TNF-a and IL-6levels on the chronic proliferative prostatitis rat.
