ObjectiveTo Investigate if the expression of claudin-1and claudin-8gene inrectal cancer cells are related,and how they work in the infestation ofthe rectal cancer.MethodsImmunohistochemical SP method was used to examineexpression of claudin-1and claudin-8gene in68cases of rectalcancer and12cases of normal cell far form cancer,the latter two wasused for contrast control.To Investigate if the expression of claudin-1and claudin-8gene in rectal cancer cells are related.RT-PCR methodwas used to examine expression of claudin-1and claudin-8gene intissue of rectal cancer,rectal adenoma and normal cell far form cancer,to confirm our assumption of their expression and correlation.ResultsThe results of immunohistochemical indicated that there werehigh expressions of claudin-1in rectal cancer(expression rate of77.9%).There was no expression of claudin-1in the normal cells farfrom cancer(expression rate of17.7%),which had significantdifference with the first two(p<0.05).There were high expressions ofclaudin-8only in few of groups in rectal cancer(expression rate of11.8%).There were high expressions of claudin-8in the normaltissues(expression rate of88.2%),which had significant differencewith the first two(p<0.05).RT-PCR method showed similar result.Theexpression strength of claudin-1was irrelevant to age and gender ofpatients,but was correlated to the degree of tumor invasion,tumordifferentiation,lymphatic metastasis and Dukes stage of rectalcancer.Claudin-8was similar to claudin-1in the characteristics ofclinical cases.ConclusionsThere were high expressions of claudin-1in rectal cancer,but not in normal tissues;while there was no expression of claudin-8in mostof rectal cancer (high expressions in some groups),but there was highexpressions in normal tissues.The high expressions of claudin-1andlow expressions of claudin-8in rectal cancer indicated thatup-regulated expression of claudin-1and down-regulated expressionwas closely related to occurrence and development of rectalcancer.The expressions of claudin-1and claudin-8inmembrane-associated protein of rectal cancer cell weredown-regulated,which may explain invasively growing and lymphaticmetastasis of rectal cancer.The high expressions of claudin-1in rectalcancer made it possible to be a tumor marker and a target for targetedtherapy.By detecting expression of claudin-1and claudin-8,we couldmeasure prognosis of patients and detect high risk group.
