| Objective To observe the sappan wood ethyl acetate extract on rat heterotopic cardiac transplantation for chronic rejection in serum IL-10levels, cardiac allograft coronary vascular changes and myocardial pathological and morphological changes of heart transplantation, for the further study of sappan wood ethyl acetate extract on chronic rejection mechanism laying foundation.Methods By using the improved Ono operation, with the male SD rats for the receptor, male Wistar rats as donors, establishment of rat heterotopic heart transplantation model in all24, after the success of the operation of continuous gavage for21days of35mg/(kg·d) cyclosporin A in the rat, were randomly divided into control group, hematoxylin group, cyclosporin A group,8rats in each group. The control group according to the weight calculation capacity of olive oil gavage; cyclosporin A group by12mg/(kg·d)cyclosporin A dissolved in olive oil administration; sappan acetic ether extract solution set to736mg/(kg·d)by gavage. Rats in each group gavage for7days after the blood collection, using ELISA assay for the detection of serum IL-10content; conventional light microscopic observation of cardiac allograft coronary arteries and heart transplantation myocardial pathological changes.Results:1.The sappan wood group, cyclosporine A group were able to make the peripheral blood IL-10levels increased significantly, compared with the control group with significant difference (P<0.05), and hematoxylin and cyclosporine group comparison, the two also have significant difference (P<0.05). And he set with cyclosporine group, there was significant difference (P<0.05).2. The sappan wood group, cyclosporine A group compared with the control group, heart transplantation myocardial was impaired to a lesser extent, a smaller area of myocardial fibrosis. Conclusion:Sappanwood ethyl acetate extract can increase the heterotopic heart transplantation rats serum IL-10content, and can reduce the transplantation myocardial pathological damage, which may be one of the reasons against chronic rejection, preliminary proof of Sappan acetic ether extract has certain immune inhibitory action. |
