| Objective: To investigate the association of several genetic polymorphisms oftransporter associated with antigen processing-1(TAP1) and predisposition to allergicrhinitis (AR) in a Uygurs and Han Chinese in Xinjiang. Methods: We conducted acase-control study in two groups, respectively with150cases of allergic rhinitis and150healthy controls in Uygurs and Han Chinese people.Three single nucleotidepolymorphisms (SNPs) rs1057141(-1332A/G)ã€rs1135216(-2245A/G)ã€rs2071480(G/T)were selected. The genetypes were determined by SNaPshot systems.And the frequenciesof genotypes and alleles of the SNPs were tested by Hardy-Weinberg equilibrium.Results:1) The distribution of TAP1*rs1057141(-1332A/G)ã€rs1135216(-2245A/G)ã€rs2071480(G/T) genetypes showed A/Aã€A/Aã€G/G are the common style, and Aã€Aã€G are the highest allelic gene in the two ethnic groups. The genetype and allelefrequencies are different in Uygurs and Han Chinese. There are five kinds haplotypes(AAG,GGG,AAT,GGT,AGT), and LD block analysis were disclosed strong LD withthree SNPs.2) In the same ethnic group:the frequencies of genetypes and alleles in HanChines case and control group showed no statistical significance (P>0.05); thedifference of TAP1*rs2071480genetypes and alleles between case and control group ofUygurs showed statistical significance (P<0.05). In the Uygurs and Han Chinese casegroup,the frequencies of genetypes and alleles were different, and in the casegroup,difference existed in the two ethnic group, and these differences showed statisticalsignificance (P<0.05).3) In the allergen skin prick test, the rate of mugwort andpigweed was the highest, but there was no significant difference between the XinjiangUygurs and Han Chinese (P>0.05).4) The serum total IgE level in AR group was higher than healthy control group, and it showed significant difference(P<0.05). Conclusion:1)The distribution of TAP1SNPs in Xinjiang Uygurs are different from other populationsin the world. In the extending research We should refer to the Uygur’s data sequencing.2)There was no different between Xinjiang Uygurs and Han Chinese in the genepolymorphisms characterization of TAP1rs1057141(-1332A/G)ã€rs1135216(-2245A/G)ã€rs2071480(G/T). Significant linkage disequilibrium was found among these three loci ofTAP1gene.3) The gene polymorphisms of rs1057141G allele plays a decisive role inthe development of AR in Xinjiang Uygurs.4) Mugwort and pigweed are the mostcommon aeroallergens with allergic rhinitis in Xinjiang Uygurs and Han Chinese.5) Theserum total IgE level in AR group was higher than control group in Xinjiang Uygurs andHan Chinese. |
