Object: Allergic rhinitis is a disease affected by both genes andenvironments with increasing prevalence by the latest years. Furthermore,allergic rhinitis devastates patient’s life quality. IL-27is a crucial cytokinein the development of inflammation,it plays an important role in allergicrhinitis. To get to know the association between IL-27single nucleotidepolymorphisms and allergic rhinitis(AR).Method: We recruited445patients with AR and691healthyvolunteers respectively in this study. DNA Purification Kit was used toextract DNA from Peripheral blood mononuclear cells (PBMCs) ofparticipators. We genotyped rs153109, rs17855750and rs181206byPCR-RFLP (polymerase chain reaction-restriction fragment lengthpolymorphism) assay.Results: There is no significant difference of age and gender betweencase group and control group. HWE (hardy-weinberg equilibrium)examination showed P﹥0.05. We found significant difference betweencase group and control group for genotypes and alleles of rs153109andrs17855750polymorphisms. In rs153109, significantly decreased frequency of CT, CC genotypes and C allele in patients unlike thatobserved in the control individuals(Pc=7.46×10-5,9.54×10-5,2.97×10-11,respectively). In rs17855750, GT genotype and G allele appeared in thepatients unlike that observed in the control individuals (Pc=1.10×10-13,4.42×10-13, respectively). while, rs153109TT genotype and T allelefrequency, rs17855750TT genotype and T allele frequency significantlyincreased than that of control group (Pc=7.72×10-10,Pc=2.97×10-11,1.10×10-13,4.42×10-13respectively). Significantly increased frequency ofhaplotypes TTC, TTT in patients with AR (P﹤0.001); while haplotypesCGT, CTC, CTT frequency significantly lower than that of control group(P﹤0.001).Conclusion: our study identified an obvious association of rs153109and rs17855750polymorphisms of IL-27with allergic rhinitis patients. |
