| Objective:This study aimed to investigate the relationship between CD14 gene single nucleotide polymorphisms (SNPs) and allergic rhinitis (AR) in Xinjiang Uygur and Han populations as well as to determine characterization of polymorphisms. Methods:32 healthy Uygurs were selected, The CD14 gene was examined by direct sequencing ensure position, types, frequency of SNPs of CD14 gene in Uygurs. To choose three high-frequency tagSNP, The case-control analysis was adopted to analyze association research. A total of 300 AR and 300 healthy controls subjects were included and their serum total immunoglobulin E (tIgE) levels were measured. Comprising 150 Uygur AR,150 Han AR,150 Uygur controls and Han controls. Screening of SNP in the CD 14 promoter was performed using SNaPshot SNP genotyping technic. The frequencies of genotypes and alleles were detected as well as the levels of tlgE in different genetypes were compared. Results:1) Eleven SNPs mutations were found in CD 14 total length in Uygurs. Two low frequency were first time to report. LD block analysis were disclosed strong LD with eleven SNPs, Among the distribution of rs2569192, rs4914, rs2569190, rs2569191 in Uygurs were quite different from Beijing Han, Japanese, European and African (P<0.05).2) The distribution of genotypes or alleles of CD 14 gene rs2569190 (-159T/C), rs2569192 (C/G), rs3138078 (-1359G/T) had no differences between the Xinjiang Uygurs and Hans (P>0.05). There four haplotypes TCG, CCT, CGG, CCG. The most frequency of alleles was T, C, G. Significant linkage disequilibrium was found among these three loci of CD14 gene.3) In the common ethnic group, the frequencies of genotypes and alleles were not different between the Han AR and Han control group (P> 0.05).The frequencies of genotypes and alleles were not different between the Uygur AR and Uygur control group (P>0.05). The frequencies of genotypes and alleles were not different between the Uygur and Han control group (P>0.05). The frequencies of genotypes and alleles rs2569192 (C/G) were different between the Uygur AR and Han AR groups (P<0.05).4) Skin prick test, The positive rate of Artemisia and pigweed were the highest (Uygurs 34.0%, Hans 39.3%). There were no significantly difference among Uygurs compared to Hans.5) The serum total IgE level in AR group was higher than healthy control group (P<0.05). Conclusion:1) The SNPs of CD14 in Uygur populations different from the others populations. We should refer to the Uygur's data in extending study.2) This study confirms CD 14 rs2569190 (-159T/C), rs2569192 (C/G), rs3138078 (-1359G/T) polymorphisms characterization were not different between the Xinjiang Uygurs and Hans. Significant linkage disequilibrium was found among these three loci of CD 14 gene.3) These three SNPs do not play a decisive role in the development of AR.4) Artemisia and pigweed are the most important aeroallergens with allergic rhinitis in Xinjiang Uygurs and Hans.5) The serum total IgE level in AR group was higher than healthy control group. No relationship between CD 14 three SNP and serum total IgE level was found (P>0.05). |
