Tissue Culture System Establishment Of Medicinal Plant Sophora Tonkinensis Gapnep

"Guangdougen" is the dried roots and rhizomes of Sophora tonkinensis Gapnep,it may also be called "Shandougen"or"Kudougen"."Guangdougen"is traditional Chinese medcine with favorable development prospects but short resources. In this study, the wild Sophora tonkinensis Gapnep were be used as the materials,it was studied on the tissue culture to explore the effective way of establishing the propagated system to provide the theoretical basis and technical support for saving the germ plasm resources and expanding the production.The results show that:1.The suitable tissue culture explant type of Sophora tonkinensis was the upper part of stem with axillary bud. the best time of getting the materials was early March,the materials had low contamination rate and high differentiation rate.2.The explants of Sophora tonkinensis were soaked in75%alcohol for30s, washed twice with sterile water, then soaked in0.1%HgCl2+tween80for10min and used the sterile water washed three times, the pollution rate was under10%.3. MS and B5were the suitable basic medium, NAA was the best auxin and6-BA was the optimal cytokinin in primary culture. The explants started to differentiate after12d in MS+6-BA1.5mg/L+NAA0.15mg/L, in30d, the differentiation rate can achieve93.33%.4. The apical-bud was the best subcultrue materials,. The addition of Coconut water in medium can improve the growth of adventitious buds. Be cultured in MS+NAA0.25mg/L+6-BA2.5mg/L+20%CW30days, the buds multiplication was3.72times.5.1/2MS medium was the optimal medium in root induction, be cultured in1/2MS+IBA1.5mg/L medium30days, the rooting rates reached91.6%,radical number was5.33,and the root length was4.56cm. the optimal transplantation method of Sophora tonkinensis was the combination of loosed lid5d+opened lid2d, and the best selection of transplantation matrices was perlite+sand+turves(1:1:1), the survival rates of seedings can reach90.00%after40days of transplant.6.The alkaloid contents of semiannual tissue cultrue seedings and seedings were different. The matrine contents in stems and leaves of seedings were0.69times,0.75times of that in tissue culture seedings. But the oxymatrine contents and total alkaloid contents of seedings were higher than that in tissue culture seedings.
