Sophora tonkinensis and Sophora tonkinensis var.polyphylla are traditional Chinese medicine in Guangxi Zhuang Autonomous Region, but 2010 Chinese pharmacopeia just embodys S. tonkinensis. In order to evaluate the quality between two kinds of herbs, their biological characteristics, microscopic structure and contents of conventional indexes was studied by conventional method, and total alkaloid were determined by the HPLC; the infrared spectral features of two kinds medicine were compared and evaluate combining with FTIR technology. The results were as follows:1. Compared with S. tonkinensis var.polyphylla, S. tonkinensis have higher height, shorter main root and lot of fibrous root. Microstructure research shows that:the roots and stems of S. tonkinensis are elliptic, but these of S. tonkinensis var.polyphylla are round; the organization of total cross section ratio of two crude drugs have much difference.2. The conventional indexes of two different collected times species were similarly. The contents of average water was 7.88% and 7.70%, respectively; total ash was 4.70% and 3.07%, respectively; alcohol-soluble extract was 16.22% and 18.41%, respectively; and their every index fit with the standard of S. tonkinensis according to 2010 Chinese pharmacopeia. Besides, the average drying rate of S. tonkinensis root was 47.40%, and the average dyring rate of S. tonkinensis var.polyphylla was 49.74%.3. The content of oxymatrine in S. tonkinensis root was 1.52%, the contents in stem and leaf were 0.28%,0.23%, respectively; the content of matrine in root was 0.20%, in stems and leaves were 0.09%,0.11%, respectively. The samples oxymatrine content was 1.4%-2.1% in different collect times, the S. tonkinensis contents was highest in July at 1.94% and in May was the lowest at 1.48%, the S. tonkinensis var polyphylla contents was highest in July at 2.09% and in March was the lowest at 1.57%. The samples matrine content was 0.15%-0.3%in different collect times, the S. tonkinensis contents was highest in September at 0.27% and in May was the lowest at 0.17%, the S. tonkinensis var.polyphylla was highest contents in January at 0.29% and in July was the lowest at 0.15%. The average contents of alkaloids were 2.02%,2.05%, respectiviy from different collection times of S. tonkinensis and S. tonkinensis var.polyphylla.4. Infrared spectrum analysis show that:two kinds of herbs contain kinds of chemical compositions, such as saccharides, protein, alkaloids, flavonoids and saponin etc. The quercetin and protein in leaf were higher than in stem and radix, relatively, but the saponin and saccharides in root, stem and leaf were similarly, so these results shows that the two kinds of herbs have similar quality.7 samples from different places were clustered in two groups in the principal component spaces, S. tonkinensis and S. tonkinensis var.polyphylla were clustered together, while Menispermum dauricum was clustered in one group alone. To some extent, the sparse density of the samples distribution reflected the genetic relationship, this further proved that there was close relationships between the two kinds of Chinese herbal medicine5. tonkinensis and S. tonkinensis var.polyphylla have similarity index in the conventional indicators, alkaloid contents and infrared spectral characteristics, except for morphology and microstructure. This text explained that two species samples was similar in quality, and also showed that it is reasonable to use S. tonkinensis var.polyphylla as S. tonkinensis according to the Guang xi Chinese materia medica standards.
