Using Tissue Microarray Of P53, Hif-1¦Á And Vegf Expression In Colorectal Cancer Tissues And Its Clinical Significance

Objective:To detect the expression of hypoxia-inducible factor 1α(HIF-1α), VEGF,P53andMVD in human colorectal carcinoma.Methods:The tissue microarray was made up of 80 cases of colorectal carcinomas and 80 cases of non-neoplasm colon mucosa tissues.The expression of HIF-1α,VEGF,P53andMVD were detected by immunohistochemistry.Results:(1)The positive rates of P53 in colorectal carcinomas was 48.7%, significantly higher than it in non-neoplasm colon mucosa tissues(P＜0.01).The expression of P53 had no relationship with the differentiation,depth of invasion,diameter of tumour(P＞0.05),but there were significant correlations with lymph-node metastasis(P＜0.01).(2) The positive rates of HIF-1αin colorectal carcinomas was51.3%,significantly higher than it in non-neoplasm colon mucosa tissues(P＜0.01). The expression of HIF-1αhad no relationship with the differentiation,depth of invasion,diameter of tumour(P＞0.05),but there were significant correlations with lymph-node metastasis(P＜0.01)(3) The positive rates of VEGF in colorectal carcinomas was 74.34%,The expression of VEGF had no relationship with the differentiation,depth of invasion,diameter of tumour(P＞0.05),but there were significant correlations with lymph-node metastasis(P＜0.01)Conclusion(1)There are overexpression of HIF-1α,P53 and VEGF in colorectal carcinoma,There were significant correlations between the expression of HIF-1 alpha,VEGFandP53 in colorectal carcinoma and progression of colorectal carcinoma(2) HIF-1 play a very important role in angiogenesis of colorectal carcinoma through regulating the expression of VEGF.(3) HIF-1 cooperates with P53 deletion or mutation taking part in the regulation of colorectal carcinomatumor cells under hypoxia probably.(4) It was feasible to study a lot of samples efficiently by using tissue microarray,for it had fast,convenient,economical and correct character.
