Objective:To investigate the expression of Galectin-3 and VEGF in gastric cancer and its significance by using microarray and immunohistochemical staining and to study their correlation with clinical-pathological features. The correlation between them,their roles in gastric cancer invasion and metastasis and the prognosis were also explored.Methods:101 cases of gastric cancer tissue sample were taken from the patients with surgical resection of gastric carcinoma in our hospital from December 2003 to December 2004. They didn't receive radiotherapy and chemotherapy before the operation and their clinical-pathological data were recorded in details. TNM staging:ΙA stage 6 cases,ΙB stage 7 cases,Ⅱstage 19 cases,ⅢA stage 31 cases,ⅢB stage 18 cases,Ⅳstage 20 cases. Normal gastric tissues from 20 patients which were gastric mucosas from the distant cutting margin(>5cm) were also accumulated as control randomly. We have made gastric carcinoma tissue microarrays. There were 20-25 cases of tumor specimens in each tissue microarray. The tissue microarrays were constructed by 4×5 or 5×5 arrangement and they are round or oval shape. Every paraffin block was sliced with 4μm thickness continuously and one slice was chosen to be dyed by HE from ten slices so that we could make sure whether the tissue was still representative. Expression of Galectin-3 and VEGF proteins were detected by using gastric tissue microarray and immunohistochemical staining in 101 cases of gastric carcinoma and 20 cases of gastric mucosas. Statistical analysis was completed by SpSS 17.0 statistic software. p<0.05 represents statistical significance.Results:1 5 paraffin blocks were prepared. The results suggested that almost all the specimens can maintain their histological types and the phenomenon of wrinkles or off-chips are rare.2 Galectin-3 is expressed in cell nucleus and cell cytoplasm,but a positive reaction is mainly distributed in cytoplasm. The positive expression rate of Galectin-3 in gastric cancer was 85.1%(86/101),significantly higher than the gastric mucosa tissue. The expressions of Galectin-3 were positively correlated with clinical stage, invasive degree, lymph node metastasis and distant metastasis(p<0.05),but was not associated with age,gender and differentiation.(p>0.05)3 VEGF is also mainly expressed in cytoplasm,only a few is distributed in cell nucleus. The positive expressions rate of VEGF in gastric cancer were 76.2% (77/101),significantly higher than the gastric mucosa tissue 25%(5/20)(p<0.05). The expressions of VEGF were positively correlated with clinical stage, invasive degree, lymph node metastasis and distant metastasis(p<0.05),but was not associated with age,gender and differentiation (p>0.05).4. Spearman correlation test show that Galectin-3 has a close relation to VEGF obviously. The expression of Galectin-3 were positively correlated with VEGF (r's= 0.4843,p<0.05).5. 12 patients were lost in follow-up because of objective reasons. 47 patients recurrent within 5 years,36 of them were died. The expression of Galectin-3 and VEGF were 97.2%(35/36) and 94.4%(34/36) respectively,significantly higher than those who have lived longer than 5 years. That suggest a lower 5-year survival rate in the patients who have the positive expression of Galectin-3 and VEGF.Conclusion:1 The tissue microarray technique has the advantage of high-throught in the detection of tumor marker,which can analyze lots of specimens and combine a variety of other biotechnology. It is also a less expense,convenient,rapid approach for screening clinical tissue specimens. It is a very valuable technique and has great prospects in early diagnosis of cancer,clinical judgement,prognosis and screening of molecular markers and so on.2 Significant correlation was found between the expresssion of Galectin-3,VEGF and the invasion and metastasis of gastric cancer.3 The expression of Galectin3 was positively correlated with VEGF,but the interaction mechanism isn't still clear. We need to do more study.4 The expression of Galectin-3 and VEGF are useful for evaluating malignancy degree,prognosis invasion and metastasis of gastric cancer.
