| [AIM]To investigate the expressions of human runt-related transcription factor 3(RUNX3),Vascular endothelial growth factor(VEGF) and microvascular density (MVD) in gastric carcinoma tissues microarray and their significance.[METHODS]Gastric carcinoma tissue microarray was prepared,and the expression of RUNX3,VEGF and MVD were detected by immunohistochemistry in 80 cases of gastric carcinoma proved pathologically.[RESULTS]1.RUNX3 expression was classified as negative 46.3%(37/80);positive 35.0%(28/80) and strong positive 18.7%(15/80) respectively,a positive reaction were mainly distributed in cytoplasm.The expression of RUNX3 in gastric carcinoma was not associated with gender,age,pathological type and Lauren's classification but was related to clinic stage(P=0.037,a=0.05).2.VEGF was strongly expressed in 47.5%(38/80);positive and negative VEGF expressions were observed in 30%(24/80) and 22.5%(18/80) respectively,a positive reaction were mainly distributed in cytoplasm.The expression of VEGF in gastric carcinoma was not associated with gender,age,pathological type and Lauren's classification but was related to clinic stage(P=0.004,a=0.05).3.MVD in expression of RUNX3 as strong positive(19.42±6.89);positive (38.21±7.63);and negative(50.13±9.28) respectively;the positive expression of RUNX3 was lower than the negative expression(19.42<38.21<50.13),(F= 64.38,P =0.00);but MVD in expression of VEGF as strong positive(51.31±8.71);positive (46.91±7.28);and negative(23.24±6.82) respectively;the positive expression of VEGF was greater than the negative expression(51.3>46.9>23.2),(F=79.83,P= 0.00).The expression of RUNX3 was negatively correlated with MVD(r_s=-0.742, P<0.01),but the expression of VEGF was positively correlated with MVD(r_s=0.683, P<O.01).4.The positive expression rates of VEGF increased gradually in negative expression (15.0<25.0%<60.0%) and weak positive(21.4%<35.7%<42.9%);but decreased gradually in strong positive of RUNX3(58.3%>33.3%>8.4%),x~2(Linear-by-Linear Association=9.178,P=0.002;the expression of RUNX3 was negatively linear correlated to VEGF(r_s = -0.333,P<0.01 ).[CONCLUSIONS]1.The expression of RUNX3 was significantly lower in gastric carcinoma,inactivation of RUNX3 was found in gastric carcinoma,and may be related to the genesis and development of gastric carcinoma.2.Increased expression of VEGF was found in gastric carcinoma,may be related to the genesis and development of gastric carcinoma.3.RUNX3 may inhibit the expression of VEGF by some mechanism and reduce the angiogenesis,growth and metastasis of gastric carcinoma.4.Combined detection of RUNX3,VEGF and MVD can estimate the biological behaviour and prognosis of gastric carcinoma patients more exactly. |
