Aim:The study is to investigate the relationship of spleen deficiency syndrom with brain-gut peptide based on the theory of five viscera holding spirit. Through observing the behavior change of the rat model of spleen deficiency syndrome, we detected immunohistochemical and genetic expression of some correlation factors of signal trasduction in order to reveal the mechanism of spleen deficiency influence on learning and memory.Methods:To study the theory, the paper set forth the importance of the Spleen Zang in the spirit, and it's mechanism. It discuss the deeper meaning of the"Yi"spirit and"Si"spirit which includes the meaning of learning and memory, and the relationship of the spleen and the learning and memory.In this study, the spleen-asthenia rat model are produced by applying multiple factors such as abnormal diet, over strain and damage from abuse diarrhea drugs. Guipi decoction was used as a remedy to verify the availability of the model. Chaihushugan decoction was used as a positive contrast. Immunohistochemical method was employed to detect the level of brain-derived neurotrophic factor(BDNF),acethlcholinesterase(AChE),nitric oxide synthase(NOS) and c-fos gene,hybridization in situ was adopted to determine the neuropeptide receptor expression of c-jun mRNA.Results:1 The spleen-asthenia rat model produced by applying multiple factors manifested symptoms similar to the typical case of spleen deficiency syndrome such as diarrhea, athrepsy, anepithymia, depressed, and so on at the thirth day. The diarrhea was severest at the eighth day. The group rat prevented and treated by Guipi decoction did not show manifestation of spleen deficiency syndrome, and compared with normal controls it did not show significant changes, which demonstrated that it was successful to make spleen-asthenia rat model.2 Immunohistochemical Detection result of BDNF,AChE and NOS1) Immunohistochemical experiment showed that the quantity of BDNF have reduced obviously in the spleen-asthenia rat models (P<0.01).However,the group rat prevented and treated by Guipi decoction did not show manifestation of reducing in the quantity of BDNF,and compared with normal group it did not show significant changes.2) Immunohistochemical experiment showed that the quantity of AChE have increased obviously in the spleen-asthenia rat models (P<0.01).However,the group rat prevented and treated by Guipi decoction did not show manifestation of increasing in the quantity of AChE,and compared with normal group it did not show significant changes. 3) Immunohistochemical experiment showed that the quantity of NOS have reduced obviously in the spleen-asthenia rat models (P<0.01).However,the group rat prevented and treated by Guipi decoction did not show manifestation of reducing in the quantity of NOS,and compared with normal group it did not show significant changes.3 Immunohistochemical Detection result of c-fos and Intrusion Detection result of c-jun mRNA1) Immunohistochemical experiment showed that the quantity of c-fos have reduced obviously in the spleen-asthenia rat models (P<0.01).However,the group rat prevented and treated by Guipi decoction did not show manifestation of reducing in the quantity of c-fos,and compared with normal group it did not show significant changes.2) Such distribution and location were also revealed in the in situ hybridization of c-jun.Conclusion:1 The Spleen Zang-Spirit theory is an important part of the Wu Zang-Spirit Theory. The Spleen Zang is the basis of the the acquired root and is the source of qi and blood production which transformation is the material foundation of spirit, and governs the pivot of qi dynamism and the water metabolizing.2 The rat model of spleen deficiency syndrome was successful which had good reduplication and reliability.3 The decreasing in the expression of BDNF,NOS,C-FOS gene,C-JUN mRNA and the increasing in the expression of AChE, correlation factors of signal trasduction of the spleen deficiency in rat's brain, clues to the mechanism of the decline of the learning and memory.4 This research is the prophase's continre, compare behavior study of Spleen Deficiency Rats ,we can concluded that correlation factors of signal transduction in the Spleen Deficiency Rats improved learning and memory of Spleen Deficiency Rats. |
