Objective JAK/STAT signal transduction pathway is an important pathway involving the signal transduction from outer cell to nucleus in the cell. This pathway regulates the ex-pression of the specific genes concerned with the processes of immunity, proliferation, differ-entiation, apoptosis and cell cycle by activating STAT (signal transducers and activators of transcription). Therefore, this study is mainly focused on the relationship between the ex-pressions of JAK1, p-STAT3 and Bcl-2 and carcinoqenesis and development of human gastric cancer. Methods Immunohistochemical methods are used to detect the expressions of JAK1, p-STAT3 and Bcl-2 in 60 gastric cancer tissues,20 chronic gastritis,20 intes-tinal metaplasia and 20 dysplasia, respectively. Results (1) The positive ratio of JAK1, p-STAT3 and Bcl-2 expression in intestinal metaplasia (30.0%,30.0% and 35.0%) and tissues adjacent to cancer (55.0%,60.0% and 53.3%) were higher than in chronic gastri-tis (10.0%,5.0% and 5.0%) (P<0.05); those in dysplasia (60.0%,50.0% and 55. 0%) was higher than in intestinal metaplasia (P<0.05); those in gastric cancer (93.3%, 93.3% and 91.7%)was significantly higher than in dysplasia and tissues adjacent to cancer (P<0.01). (2) In gastric cancers the positive intensities of JAK1, p-STAT3 and Bcl-2 expression were not correlated with age, sex and tumor size (P>0.05), but associated with differentiations, depth of tumor invasion, lymph node metastasis, distal metastasis and clini-cal stages (P<0.05); (3)Overexpression intensities pencents of JAK1, p-STAT3 and Bcl-2 protein in poorly differentiated gastric cancer (71.9%,66.7% and 67.7%)were higher than in highly and moderately differentiated ones(25.0%,34.8% and 20.8%)(P<0.01); those in gastric cancer with an invasion depth to the outer serosa(77.8%,78.6%and 80. 8%)were higher than with an invasion depth to inner serosa(27.6%,25.0%and 17.2%) (P<0.01); those in gastric cancer with lymph node metastasis(58.3%,60.4%and 52. 2%)were higher than without lymph node metastasis(12.5%,12.5%and 22.2%)(P<0. 05);those in gastric cancer with distal metastasis(79.2%,78.3%and 81.8%)were high-er than without distal metastasis(31.3%,36.4%and 24.2%)(P<0.01);those in gastric cancer of stageⅢ-Ⅳ(80.8%,73.1%and 82.6%)were higher than in gastric cancer of stageⅠ-Ⅱ(26.7%,36.7%and 20.6%)(P<0.01);(4)Every two expressions of JAKl,p-STAT3 and Bcl-2 in gastric cancer were compared can be show correlation.γs (0.2846,0.4595,0.2905)>γs0.05(60)=0.255,(P<0.05).ConcIusion The JAK1,p-STAT3 and Bcl-2 were overexpressed in gastric cancer tissues compared with normal gas-tric tissues;The overexpression of JAK1,p-STAT3 and Bcl-2 in gastric cancer probably relate to its oncogenesis,invasion,metastasis and clinical stages closely;The expressions of JAK1,p-STAT3 and Bcl-2 in gastric cancer had correlation.
