Objective: To explore the mechanism of Huazhuojiedu Decoction through observating its’ effect on the expression of Janus kinase 1(JAK1),signal transducer and transcriptional activator(STAT3),and claudin-1 in the colon tissues of UC rats.Methods: 40 8-week old male Wistar rats were randomly divided into two groups according to random number table method :10 rats were put into the normal group,30 rats were used to build model rats by 3.5% dextran sulfate sodium(DSS).After the completion of the building model,the 30 rats were further divided into three groups according to random number table method,namely: the model group(MG),Mesalazine group(XG)and Huazhuojiedu Decoction group(ZG),each group of nine.NG and MG were given normal saline by gavage,XG and ZG were given corresponding drugs by gavage,and the changes of general mental state,food intake,activity,stool and body weight of the rats were closely observed every day to evaluate the Disease Active Index(DAI).After 14 days of treatment,the rats were sacrificed and their colon tissues were taken.The pathological changes of the colon tissues were observed by HE staining,and the protein expressions of JAK1,STAT3 and claudon-1 in the colon tissues of the rats were detected by western-blot method.Results:1.General situation and DAI scoreAfter DSS modeling,the rats in the MG were small in body,dull in hair,listless in spirit,less active in curl,and slow in reaction.Their food intake was significantly reduced,their weight was reduced,and their symptoms of mucous pus,blood and stools were significantly aggravated.After 14 days of intragastric administration,the above symptoms in the XG and the ZG were significantly improved,while the overall symptom improvement in the model group was less obvious.The results of DAI scores showed that compared with NG,the DAI scores of MG,XG and ZG rats were significantly increased with statistically significant difference(P<0.05),while compared with MG,the DAI scores of XG and ZG rats were significantly decreased with statistically significant difference(P<0.05).There was no statistically significant difference in DAI score between ZG and XG(P>0.05).2.The mucosal manifestations of the colonic tissues of rats in each group were observed with the naked eye and under the microscopeMacroscopic observation showed that the surface of the colon mucosa in MG was rough,with hyperemia,edema,erosion and ulceration visible.The situation was the worst,while the mucosa of XG and ZG were smooth,without obvious hyperemia,edema,erosion and ulceration,and there was no obvious difference between the two groups.The microscopic observation,the structure of colonic mucosa in MG were damaged,the local mucosa layer was damaged,the gland shape was irregular and scattered,crypt abscesses,goblet cells decreased,inflammatory cells infiltration,and rats colonic mucos damage in XG and ZG was not obvious,crypt abscess disappeared,inflammatory cells reduced,goblet cells increased,histopathologic improved.3.Results of protein detection by western-blot3.1 The expression level of JAK1 protein in colon tissues of each groupCompared with NG,the expression level of JAK1 protein in the colonic tissues of other groups was increased,and the difference was statistically significant(P<0.05).Compared with MG,the expression level of JAK1 protein in the colonic tissues of rats in XG and ZG was decreased,and the difference was statistically significant(P<0.05).Compared with ZG,the expression in XG was lower,and the difference was statistically significant(P<0.05).3.2 The expression level of STAT3 protein in colon tissues of each groupCompared with NG,the expression level of STAT3 protein in the colonic tissues of the other groups increased,and the difference was statistically significant(P<0.05).Compared with the MG,the expression level of STAT3 protein in the colon tissues of rats in XG and ZG decreased,and the difference was statistically significant(P<0.05).Compared with ZG,the expression in XG was lower,and the difference was statistically significant(P<0.05).3.3 The expression level of claudin-1 protein in colon tissues of each groupCompared with NG,the expression level of claudin-1 protein in the colonic tissues of other groups was decreased,and the difference was statistically significant(P<0.05).Compared with MG,the expression level of claudin-1 protein in the colon tissues of rats in XG and ZG was increased,and the difference was statistically significant(P<0.05).There was no significant difference between ZG and XG(P>0.05).Conclusion:(1)Huazhuojiedu Decoction has a good therapeutic effect on DSS-induced ulcerative colitis model rats,which can improve its general state,repair the pathological damage of colon tissues,and promote the healing of ulcer.(2)Huazhuojiedu Decoction may protect the intestinal mucosal barrier and repair the damaged intestinal mucosa of UC rats by inhibiting the JAK/STAT pathway and regulating the expression of tightly bound claudin-1. |
