| Objective:1.The establishment of a viable,good repeatability,and the high rate of successful operation of arterial reconstruction rat model of orthotopic liver transplantation for liver transplantation research after transplantation immunology, biliary complications to provide a more consistent with the physiological animal model 2.Research on the model of postoperative biliary pathology,biochemical changes.3.Observed arterialized on rat liver transplantation for the protective effect of bile duct.Method:1.Using male SD rats from the same strain of allogeneic transplantation for liver receptor.Donor surgery:block cut the liver donor,and will be in the preservation of donor portal for liver and inferior vena cava put under the cuff; receptor surgery:to the classic "two-cuff" basis and then to the donor celiac artery and the right renal artery for-receptor - to-end anastomosis,the establishment of the hepatic artery reconstruction of rat liver transplantation model2,from 64 male SD rats with a strain of rat liver transplantation for the receptor,and were divided into two groups,non-arterial double-cuff technique rat liver transplant team as a control group, artery double-cuff technique Rat Liver Transplantation Unit,as the experimental group as a control group.Control group and experimental group after modeling success,each 24,each time point(after 1,2,4,7 days)each for six anesthesia inferior vena cava blood,from bile duct,liver,respectively function and histopathological examination.Results:1.Succeeded in establishing a double-cuff technique artery of the rat liver transplantation model,a success rate of 92.8%,87.3%for the week survival.The average operative time:Donor(50±10 min),sleeve anastomosis(2±1 min),in vitro repair time for the liver(11±1 min),receptors(35±10 min),vascular anastomosis (15±10 min),no warm ischemia time,cold ischemia time(45±10 min),the receptor liver(15±2 min).40 cases of formal experiment,in which three cases of death,and one of them died of respiratory arrest,2 patients died of liver upper and lower vena cava anastomotic bleeding.37 rats survived in the two cases,one case of the first three days after the death of death was stomach expansion;another one cases in the first five days after the death,the reasons for the link for biliary stent Weida leakage of bile formation peritonitis.After one week,the rats were sacrificed not found in celiac artery embolization.2,the experimental group and the control group,weight,surgical success rate,one week survival no significant difference(P>0.05)in postoperative liver function,the experimental group was significantly better than the control group(P<0.05),after the big Rat ALT,Tbil detected artery model of the arteries than non-significant difference,r-GT a significant difference(P<0.05).In pathology,and the experimenters compared to the control group,in the first postoperative day and the two days there was no significant difference from the beginning of the fourth day after, can be observed that the control group bile duct epithelial cells exfoliated partial necrosis,the 7th postoperative days can be observed biliary epithelial cells are disordered,and even degeneration,and the experimental group biliary epithelial damage significantly lighter than the control group.Conclusion:1.we have established a double-cuff technique artery of rat liver transplantation model is simple,repeatable,and the success rate higher.2.arterial model can maximize the protection of the liver histological structure can significantly reduce the incidence of biliary complications and improve long-term survival. |
