Objective: Establishing a rat model of orthotopic liver transplantationby two-cuff technique, the purpose is to study the immune tolerance.Methods: There are220Sprague Dawley rats and20Wistar rats were treated with liver transplantation by improved two-cuff method.The first and second stage line SD-SD50cases of liver transplantation, respectively. The third stage established rat liver allograft rejection (Wistar-SD) model20cases. Record survival rate after24hours, the operation time of the donor, reparative time of donor liver, the operation time of the recipient, hepatic warm ischemia time, hepatic cold ischemia time, anhepatic phase and supra-hepatic inferior vena cavareconstruction time. Analysing the main reason for surgical failure.Results: Establishment of SD-SD100cases and Wistar-SD20casesof liver transplantation model, Phase124hours survival rate was0; Phase2was20%; Phase3was55%; The main reasons for the death ofintraoperative were bleeding, anesthesia accidents, poor quality of the liverand so on. The main reasons of death after operation were bleeding,anastomotic leak, no function of liver, thrombosis and biliary complications etc. Phase3donor and recipient operative time, reparative time of donorliver, hepatic cold ischemia time, anhepatic phase and supra-hepaticinferior vena cava reconstruction time shorter than the first and secondstage, there are significant statistically difference(P<0.05), and a gradualincrease survival rate after surgery.Conclusion: In order to successfully establish a rat model oforthotopic liver transplantation, based on lots of rat liver transplantationtraining, requires skilled microsurgical techniques, improve microvascularsuture technique and reduce various complications can get a high survivalrate and stable rat model of orthotopic liver transplantation. |
