Object: To establish ulcerative colitis model in rats by 'the compound method of immune induction in combination with chemical stimulation', study and discuss the influences of abnormality of expression of Fas, FasL and Bcl-2 in epithelial cells of colon mucous membrane and in lymphocytes of colon tissue together with imbalance of cytokine network in pathogenesis of ulcerative colitis, and observe the therapeutic effect of 'kuijiefang' decoction on ulcerative colitis rats and its influences on abnormality of cellular apoptosis and imbalance of cytokine network.Method: 36 male wistar rats were divided into 6 groups at random: Group B, M, C, L and I. H. Ulcerative colitis model was established by 'the compound method of immune induction in combination with chemical stimulation' in rats. The pathological changes of colon tissue and general conditions (including stool, weight and status of activity) were observed and evaluated. The different groups were given different disposal: administer 1.2%SASP to rats of Group C, administer 'kuijiefang' decoction in different dose (12%, 68% and 124% respectively) to rats of Group L, I, H respectively, and administer equal quantity of normal saline to rats of Group B and M, 2.5ml/100g weight, intragastrically. The samples of colon tissue and plasma from rats were taken at the end of the test to observe the pathological changes of colon tissue and make evaluation, the expression of Fas, FasL, Bcl-2 in epithelial cells of colon mucous membrane and in lymphocytes of colon tissue, and the plasma level of TNF-a, and the colon tissue level of IL-10. The differences of indications between different groups were compared and then analyzed by statistical method.Results:Comparing with Group B, the rats of Group M presented diarrhea swiftly after establishment of ulcerative colitis models, diarrhea lasted for a comparatively long time. bloody stools, mucous stools and pus stools might be found in some rats. The weight and activity decreased, the hair of the rats became tousled and lost its luster. The colon tissue of UC rats presented ulcerative colitis-like pathological changes. After administering 'kuijiefang' decoction, those abnormal presentations, mentioned above, of general conditions and the pathological changes in colon tissue were improved considerably. The middle and high dosage of 'kuijiefang' decoction had better effects, their results were close to SASP and similar to that of Group B.In Group M the expression of Fas, FasL, Bcl-2 in epithelial cells of colon mucous membrane and in lymphocytes of colon tissue increased, and there was phenomenon of abnormal expression at aberrant position in epithelial cells. 'kuijiefang' decoction exert significant function on correcting these abnormalities. The middle and high dosage had better effects, but not in low dosage.In Group M, the level of colon tissue IL-10 decreased and the level of TNF-a increased. 'kuijiefang' decoction could return these abnormalities to their normal conditions respectively. The effects of 'kuijiefang' decoction in middle and high dosage were more considerable, their results were close to SASP and similar to Group B. The effect of 'kuijiefang' decoction in low dosage was not considerable.Conclusion:'The compound method of immune induction in combination with chemical stimulation' can be used to establish ulcerative colitis in rats successfully. The model can simulate general conditions and pathological changes of human ulcerative colitis so that it can be used in pharmacological research and test dealing with ulcerative colitis.'kuijiefang' decoction can exert therapeutic function on ulcerative colitis in rats. The 'kuijiefang' decoction in middle and high dosage have comparatively better effects, unfortunately, this research failed to find the relationship between the effect and the dosage of decoction.The mechanism of 'kuijiefang' decoction in curing ulcerative colitis in rats may involve aspects as follows: return the abnormal expression of Fas, FasL, Bcl-2 in epithelial cells of colon mucous membrane and in lymphocytes of colon tissue to the normal conditions, and decrease the plasma level of TNF-a, increase the tissue level of IL-10. Thereby, it is considered that the mechanism of 'kuijiefang' decoction in curing ulcerative colitis bears relationship with its function of correcting the abnormalities in cellular apoptosis and re-establishing imbalance of cytokine network. |
