ChangAnKang Capsule is a clinical experienced prescription which is composed of Huangqi, Jinyinhua, Diyu, Huanglian, Buguzhi, Weimuxiang. It has the effect of Jianpi, Yiqi, Qingre, Zaoshi, Huoxuehuayu et al, and is mainly used in treating diarrhea et al. This project aims at testifying the effect of ChangAnKang on anti-experimental UC and further evaluates the mechanism of its effect.Pharmacodynamic Study of ChangAnKang in Treating UC 1 The protective effects of ChangAnKang on experimental UC rats 1.1 Effects of ChangAnKang on UC rats induced by acetic acidThe ulcerative colitis rat models were induced by acetic acid enema, treated with ChangAnKang and SASP and 05 %CMC,then observed the general situation and colonic injury, and checked the change of MPO and free radicals in colonic tissue. These rats evidently evacuated watery stool, they curled up and rarely moved, unanimated, hair-erected, back-humped, dirty fur, with bad appetite, and the result of Occult-Blood Testing were strong positive (-H-+).After the treatment, these rats somewhat recovered: they had better appetite and seldom watery stool, and the result of Occult-Blood Testing were between positive (++) and weak positive (+)After the treatment, the following were observed: the three dosages of ChangAnKang(1.0g/kg, 0.5g/kg, and 0.25g/kg)had no obvious effect on the experimental rats' weight (P>0.05); all three dosages had obvious improvement on the rats' swollen colon (P<0.05); the dosages l.Og/kg and 0.5g/kg had very obvious positive effect on the curtate colon (P<0.01); all three dosages improved very obviously on the broaden colon (P<0.01); all improved the colonic injury; all had very evidently effect on reducing the rising activity of MPO (P<0.01), and had effect on lowering the NO in the tissue, and lowering, to some extent, the rising activity of NOS.A11 these dosages can evidently increase the depressed activity of these rats' SOD (P<005), and at the same time, can evidently decrease the rats' MDA (in particular the two dosages 1.0g/kg and 0.5g/kg) (P<0.05). The three dosages also had obvious effect on increasing the activity of GSH-PX (P<0.01). Histopathological examination showed that all three dosages had evidently inhibited inflammatory infiltration, reduced the ulcered region, repaired the epithelia, proliferated the granulation tissue, and accelerated heal up of the ulcer. 1.2. Effects of ChangAnKang on UC rats induced by TNBSAfter preparation of TNBS-induced UC rat models, the animals were divided into 6 groups and were treated with, respectively, normal saline, the three dosages of ChangAnKang (l.0g/kg, 0.5g/kg, and 0.25g/kg), and SASP, observed the general situation of the rats, damage index of the colon and the inflammation. The result showed that, on the second day, the rats evidentlyevacuated watery stool (blooded stool were observed even by the naked eyes), they curled up and rarely moved, unanimated, with bad appetite, and the result of Occult-Blood Testing were strong positive (+++). Some died and anatomy showed that the colon were either perforated or highly conglutinatedAiter the corresponding treatment, the treated groups of rat recovered quickly: they were lively, watery stool were less or simply disappeared, and the results of Occult-Blood Testing were between positive (++) and weak positive (+). Then the rats were put to death and it showed that, all three dosages of ChangAnKang had no obvious effects either on the losed weight of the rats or on the curtated colon (P>0.05y, all have positive effects on the broaden colon (PO.05); all can improve the general damage index (P>0.05); all can improve the colon pathomorphology, in particular the l.Og/kg and 0.25g/kg dosages; as for anti-inflammation, all three dosages decreased evidently the activity of MPO (PO.01), this showed that ChangAnKang had obvious the effects on anti-inflammation.Conclusion: ChangAnKang Capsule had obvious protective effects on experimental colonitis, and the main reasons for those effects perhaps lie in its anti-inflammation and anti-free radical effects.2.
