Objective: Primary liver cancer has below characteristics : high occurrence rate in the world, there are about more 100 millonal new sufferers every year, high death rate in China. It's essential characteristic is invasion and metastasis what the key cause brings to dead . So it's very important to protect from invasion and metastasis.The Traditional Chinese medicine has accumulated abundant experience of the treatment of liver cancer. According to the Tradional Chinese medicine theories and practice of curing this kind of disease, we developed this compound prescription JiaYiJian. It also reflects Wang Xugao's and Qin Bowei's experience of using Chinese herbal medicine. According to the clinical application, the curative effect was remarkable. The aim of our experiment was to study the mechanism of anti-invasoin and metastasis of JiaYiJian in mice with transplanted liver cancer(H22), especially to observe the effect of JiaYiJian on E-cadherin and CD54 expression in mice hepatocellular carcinoma tissue. To provide experiment data, to produce the scientific evidence for further guiding and extending the clinical application.Methods: 75 male mice (SPF) were transplanted liver cancer (H22)subcutaneously into left footpads to be established as experimental lymphatic metastasis models, and were divided randomly into 5 groups (each group had 15 mice): the patholiogic model group (group A), the westen medicine group (group B), JiaYiJian low, middle, high dose treatment groups (group C,D,E). From the next day , the groups were treated by JiaYiJian low, middle, high dose and 5-Fu respectively,the pathological group was given the same dose NS (Normal Saline), Once everyday. After 20 days, the mice were killed and weighed.The thymuses, spleens, and their left footpads were excavated, weighed, and some parts of them were performed on parafomaldehyde-fixed, paraffin-embedded secions. Count the index of thymuses, spleens. The expression of E-cadherin, CD54 were also measured with immunohistochmeistry staining.Results: 1 The comparisons of the index of spleens among different groups: The group B is significantly lower than the group A, C, D, E, shown on statistics with significant difference (P<0.01), No significant difference shows between the group A and the group C, D(P>0.05),The group A is lower than the group E, with significant difference(P<0.05), the group C, D are lower than the group E, with significant difference(P<0.01 or P<0.05), No significant difference shows between the group C and the group D(P>0.05). 2 The comparisons of the index of thymuses among different groups: The group A is lower than the group D, E, with significant difference(P<0.05), No significant difference shows between the group A and the group B,C(P>0.05),No significant difference shows among the group B, C, D,E(P>0.05).3 The comparisons of the invasion of hepatocellular carcinoma tissue among different groups: all treatment groups are lower than the group A ,with significant difference (P<0.01), No significant difference shows among all treatment groups (P>0.05). 4 The comparison of E-cadherin expression among five groups: the group A is lower than all treatment groups, with significant difference(P<0.01 or P<0.05), No significant difference shows among all treatment groups (P>0.05). 5 The comparison of CD54 expression among five groups: all treatment groups are lower than the group A ,with significant difference (P<0.01 or P<0.05), No significant difference shows among all treatment groups (P>0.05).Conclusion: 1 The JiaYiJian can increase mice immune activity and depends on the dose of JiaYiJian in proper range. The group E has the most significant effect on the index of thymuses, spleens. 2 JiaYiJian can inhibit the invasion of hepatocellular carcinoma tissue. 3 JiaYiJian can increase E-cadherin expression in hepatocellular carcinoma tissue. 4 JiaYiJian can decrease CD54 expression in hepatocellular carcinoma tissue. Its mechanism may is to improve immune functions and injury the carcinoma cell, to increase E-cadherin and decrease CD54 in liver cancer ,so as to inhibit the tumor angiogenesis.
