Association Analysis Between The Norepinephrine Transporter Gene, Social Psychic Factors, Their Interaction And Major Depression

Objective:1.To analysis the association between NET gene polymorphism(including T-182C and G1287A)and Major Depresson(MD).2.To analysis the association between social psychic factors,their interaction and MD.3.To analysis the interaction between NET gene polymorphism and Social psychic factors in MD.Methods:A case-control sample,including 281 patients and 281 controls,was recruited in our study with strict selected standards.HAMD,EPQ,LES,SSS and SCSQ were used to evaluate the Social-psychic factors,such as personality,life events,social supports and coping styles. Polymerase chain reaction(PCR)and DNA direct sequencing technique were used to detect the T-182C and G1287A genetic polymorphism of the NET gene.All statistical analysis was performed using SPSS statistical software.The genotype and allele frequency distribution were compared with X~2 analysis.Haplotype analysis was performed using UNPHASE Software,and multivariate logistic regression were used to screening Social-psychic factors and analysis the interaction between NET gene polymorphism and Social-psychic factors in MD.Results:1.There was no significant difference of genotype frequency of NETT-182C between case and controls(P=0.128).The same result was observed in NET G1287A(P=0.513).But the T-182C C/C was probably association with MD,referring to T/T,its OR was 1.844(P=0.046).2.Haplotype analysis showed that the frequency of haplotype A-C in case was significant higher than control(P=0.000),its OR was 2.759(1.771-4.296).There was no significant difference of frequency of haplotype A-T,G-T or G-C between case and controls(P＞0.05).3.The result of association analysis between social psychic factors,their interaction and MD3.1 The result of multivariate logistic regression showed that Introverts,Neuroticism,LES, Negative coping style,low-Positive coping style were the related Social-psychic factors of MD. Their OR(95%CI)respectively were 4.508(2.774-7.328,P＜0.01),7.514(4.328-13.044,P＜0.01),2.517(1.535-4.126,P＜0.01),2.955(1.490-5.861,P=0.002),4.513(2.637-7.722, P=0.009).3.2 The result of interaction of Social-psychic factors:There were significant interaction among Psychoticism and Introverts,Neuroticism,Negative coping style,low-Positive coping style,low-social support,LES;There were significant interaction among Introverts and Neuroticism,Negative coping style,low-Positive coping style,low-social support,LES;There were significant interaction among Neuroticism and Negative coping style,low-Positive coping style,low-social support,LES;There were significant interaction among LES and Negative coping style,low-Positive coping style,low-social support;There were significant interaction among low-social support and Negative coping style,low-Positive coping style.4.The result of interaction of NET gene polymorphism and Social-psychic factor:4.1 The result of interaction of NET T-182C and Social-psychic factor showed that there were significant interaction among genotype C/C and Psychoticism,Neuroticism,LES,Negative coping style,low-Positive coping style and low-social support.There were not interaction between genotype C/C and introverts.4.2 The result of interaction of NET G1287A and Social-psychic factor showed that there was significant interaction among genotype A/A and Psychoticism,Neuroticism,LES,Negative coping style,low-Positive coping style and low-social support..There was no significant interaction between genotype A/A and Negative coping style,and low-Social support.Conclusions:1.T-182C C/C was associated with MD,G1287A gene polymorphism was not associated with MD.2.The haplotype A-C may increase the risk of MD.3.Neuroticism,Introverts,LES,Negative coping style were probable associated with MD,they may increase the risk of MD;Positive coping style and Social support may depress the risk of MD.There were significant interaction among Social-psychic factors.4.There were significant interaction among genotype C/C and Psychoticism,Neuroticism, LES,Negative coping style,low-Positive coping style and low-social support.There were not interaction between genotype C/C and introverts;There were significant interaction among genotype A/A and Psychoticism,Neuroticism,LES,Negative coping style,low-Positive coping style and low-social support.There was no significant interaction between genotype A/A and Negative coping style,and low-Social support.
