Objective:1. To validate the influence of social psychic factors during the association study between serotonin transporter (5-HTT) gene polymorphism and major depression (MD).2. To investigate the relationship between 5-HTT gene polymorphism (including 5-HTTLPR and Stin2) and morbility, severity and suicidal idea of MD under the controlling of social psychic factors,Methods:A case-control sample, including 152 patients and 152 controls, was recruited in our study with strict selected standards. HAMD, EPQ, LES, SSS and CSQ were used to evaluate disease serious degree and the social psychic factors, such as personality, life events, social supports and coping styles. PCR was used to detect the polymorphism of 5-HTTLPR and Stin2. The genotype and allele frequency distribution were compared withχ2 analysis. Comparison of means was used with ANCOVA, and multivariate logistic regression was used to control social psychic factors and compute odds ratio.Results:1. After controlling the social psychic factors, the OR values of two polymorphism were all proceed some change, which hint that social psychic factors influenced relevance between 5-HTT gene polymorphism and MD.2. Results of revelence between 5-HTTLPR gene polymorphism and MD①There was a significant difference of genotype frequency of 5-HTTLPR between patients and controls (P<0.05). The frequency of LS genotype in MD group (33.6%) was higher than control (19.1%) and the OR value was 3.670 (95%CI=1.468-9.177, P=0.005). There was no statistical difference of allele frequency between two groups.②Taking social psychic factors as covariant, the comparison of HAMD score between 5-HTTLPR genotypes and alleles had no significant difference. There were no significant differences of the distribution of genotypes and alleles between two HAMD grades, and the OR values were not get statistical significance yet.③There were no significant differences of the distribution of 5-HTTLPR genotypes and alleles between suicidal idea group and non-suicidal idea group, and the OR values were not get statistical significance yet.3.
