Objective To observe the effect of electro-acupuncture(EA) of different frequency on affecting ability of spatial learning and memorizing in rats with deficiency of learning and memorizing. To select the best frequency of EA for improving spatial learning and memorizing in rats with deficiency of learning and memorizing. Meanwhile observe the intervention of EA in the content of excitatory amino acid and protein expression of NR2B subunit in rats, so as to discuss the mechanism of EA on affecting learning and memorizing in rats.Methods The model rats with deficiency of learning and memorizing chose by Morris water maze were divided into one blank control group and four EA therapeutic groups including the 2Hz group, the 15Hz group, the 100Hz group and the 2/100Hz group. Needled Baihui and Yamen acupoints in rats with the electro-acupuncture once a day and remained the needles for 30min per time, gave a continuous course treatment for 18 days. We observed the spatial learning and memorizing in all rats by Morris water maze, detected excitatory amino acid content by automatic amino acid analyzer and protein expressionof NR2B subunit in brain tissue with immunoblotting in all rats.Results First, the spatial learning in all EA therapeutic groups was improved.The improvement in the 2Hz group was better than the others (P<0.05).
