Objective:The research used electro-acupuncture as a treatment tool, and observed the changes of learning and memory abilities of VD modeled rats with Morris water maze, and then, to find the effects of electro-acupuncture on SCN clock, Bmall genes and CA1ERK gene. Thus to detect the inner clock regulating mechanism of electro-acupuncture effects on VD modeled rats which would provided a scientific basis for the clinical application of treatment of VD by electro-acupuncture.Methods:1. To detect effects of electro-acupuncture on learning and memory abilities of2-VO modeled rats with the Morris water maze. And to discover the differences between western medicine group, model group, sham-operationgroup and electro-acupuncturegroup.2. To detect expression of SCN clock, Bmall mRNA and ERK mRNA in hippocampus CA1region of each group, and the differences between groups.Results:1. The Morris Water Maze behavioral test of VD-model-based rats showed that positive drug group and electro-acupuncture group d4, d5had significant difference (p <0.05) compared to the model group. As to the sham operation group, d4had significant difference while d5had not. Space exploration test analysis showed that electro-acupuncture group (p<0.01), positive drug group (p<0.05), and sham operation group (p<0.01), had significant difference on the frequency of rats into three quadrant.2. Quantitative PCR electrophoresis showed the PCR product of SCN clock gene, Bmall gene and hippocampal CA1ERK gene were only a specific single band, and the size of the line was the expected results; optimize the reaction conditions of each gene, got the best reaction conditions and the system;.3. Electro-acupuncture on Baihui and Shenmen could elevate SCN clock gene expression of rats (P<0.05) and expression of SCN gene, Bmall gene, hippocampal CA1ERK1gene and ERK2gene had improved trend.Conclusions:1. In space exploration task, electro-acupuncture and drug group had positive improvement trend. Thus, electro-acupuncture could improve the learning and memory ability of VD rats which were ischemia. And the paper provided reference to a certain extent for clinicians to treat ischemia-induced VD patients.2. The electro-acupuncture group, same with positive drug group and sham operation group, showed improvement trend. Although the expression of mRNA had a time delay to the effect on behavior, the use of real-time PCR to detect the expression of the corresponding genes can indeed accurately reflect the pathological changes and the therapeutic effect of electro-acupuncture on VD rats.
