Objective To ascertain if the apolipoprotein E (apoE) gene polymorphism is associated with β amyloid protein and vascular dementia(VD).Methods The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect apoE genetype in 25 VD patients, 25A D patients and 25 healthy age-matched controls. Meanwhile, The expression of β amyloid gene in peripheral mononuclear blood cell(PMBC) was examined using quantitive competitive RT-PCR methods .Results The increased frenquency of apoE 4 allele (P<0.01) and decreased apoE e 3 allele frenquency (P<0.05) were found in both VD and AD . There was no difference between the two groups. In every ug cDNA, the expression of β amyloid in PMBC were (3.58 0.90)amole ,(3.05 0.87)amole and(2.31 0.56)amole respectively.Obviously, the 3 amyloid was overexpressed in AD compared with health control(t=5.96,P<0.01) or VD cases(t=2.09,P<0.05); the 3 amyloid was overexpressed in VD compared with health control(t=3.57,P<0.01); In every group, the expression of 3 amyloid among different genotype groups were different, which in e 4 carrier group were higher than in E 2 and e 3 carrier group,but there is no significant influence. Conclusions apoE polymophysims may be associated with vascular dementia. apoE 4 allele could probably be a predisposing risk mark for the disease.The expression of 3 amyloid inPMBC can not be significantly influenced by the apoE e 4 allele in AD, VD and control group.
