Objective:To observe the effects of different dose of ShenQi compound recipe (SQCR) on ox-LDL、8-iso-PGF2a、3-NT of type 2 diabetic macroangiopathy KKAy mice. And to provide guidance for clinical therapeutic usage, taking oxidative stress as starting point to explore the action mechanism of SQCP in type 2 diabetic macroangiopathy.Methods:For adapting themselves with the surroundings, keep 115 2-months old KK-Ay mice of spontaneous type 2 diabetic, whose fasting blood-glucose≥ 11.1mmol/L, for 1 week.The sex of these mice is male. Randomly divided them into 6 groups indicated no statistic differences by average weight in group and between groups,25 for control group,30 for model group,15 for ShenQi compound recipe-high dosage group (SQCRH),15 for ShenQi compound recipe-low dosage group (SQCRL),15 for ShenQi compound recipe-middle dosage group (SQCRM),15 for Metformine group. The mice in control group are fed normally. Mice in the other five are continuously fed high fat for 8 weeks to establish Metabolic Memory of Hyperglycemia with Diabetic Macroangiopathy model, then medicine intervention therapy from week nine. Control group and model group were given normal saline, 10ml/kg·d. SQCRL were given ShenQi compound recipe extract 7.2g/kg·d. SQCRM were given ShenQi compound recipe extract 14.4g/kg·d, SQCRH were given ShenQi compound recipe extract 28.8g/kg · d, Metformine group were given Metformine suspension liquid,0.167g/kg·d. To observe general situation of each group during the experiment, taking 10 mice each from control group and model group to detect pathology、INS、blood lipid and oxidative stress to check modeling in medicine intervention therapy week 0. Kill all the mice at gavage week 6, testing index of fasting blood-glucose、blood lipid、INS、oxidized low-density lipoprotein of serum (ox-LDL)、3-nitrotyrosine (3-NT)、8-iso-prostaglandin-F2a (8-iso-PGF2 a) and the change of the pathology section of KK-Ay mice’s aorta, then analyze differences of index among groups by statistical software.Results:Life state、fasting blood-glucose、blood lipid、INS、oxidative stress of SQCRM group and KK-Ay group decreased compared with model group. HE Staining in thoracic aortic indicated that the lesion of SQCRM group and KK-Ay group is lighter than model group.The effect of SQCRM is better than SQCRH group and SQCRL group.Conclusion:①High fasting blood-glucose and high blood lipid and Hyperglycemia with Diabetic Macroangiopathy of KK-Ay mice who have the genes of obese and Type 2 Diabetes Mellitus can be effectively ameliorated by changing diet structure into low fat.②The mental state、weigh、blood lipid、fasting blood-glucose and the index of ox-LDL、3-NT、8-iso-PGF2 a of KK-Ay mice can be removed by giving full dose SQCR in the early time. We can infer that SQCR can light the hurts wich made by oxidative stree to the aorta of KK-Ay mice by multiple target pionts and paths.③ Middle dose is the best dosage of SQCR,because of middle dose can keep all index at a level of well and stable.The overall effect is better than low dose and high dose.
