Objective:(1)To observe the effect of Shenqi compound prescription(SQCP) inhibiting serum ox-LDL、8-iso-pgf2a、and3-NT of type2diabetic macroangiopathy KKAy mice in different treating points;(2)To discuss the mechanism of SQCP improving diabetic macroangiopathy from the perspective of oxidative stress to guide the clinical medication to some extent.Method:The1887-8weeks old male KKAy mice are kept with no intervention for a week to adapt themselves, and then randomly divied into four groups:normal group52, model group53, Chinese medicine group(Shenqi compound prescription)43, Western medicine group(Metformine)40. Mice in the normal group are feed ordinarily, and the rest of the three with high-fat diet for8weeks to copy the spontaneous type2diabetic macroangiopathy mode. Drug administration:mice in both the normal group and model group are drenched with normal saline on the criteria of10ml/kg.d, mice in Chinese medicine group are drenched with SQCP extract on the criteria of14.4g/kgd, while mice in Western medicine group with Metformine suspension according to0.167g/kgd. Index selection:after8weeks’ high-fat diet10mice are randomly chosen from the normal group and model group respectively for the detection of pathology%blood lipid and oxidative stress to ensure a successful model-building; About10mice from the four groups are chosen to death in the3th、6th and9th week respectively to obtain fasting blood samples for the detection ofTC、TG、HDL-c with biochemical method and ox-LDL、8-iso-PGF2a、3-NT with ELISA method.Results:Mice with high-fat diet show obvious abnormality in those index above.For the mice in the Chinese medicine group when compared to the model group,general life state and blood glucose are improved and relived;serum TC、TG levels drop significantly while HDL-c rises,; serum levels of ox-LDL、8-iso-PGF2a and3-NT drop significantly, all of which show certain time-effect relationship:it tends to take effct at the3th week of administration and the effect gets further improved in6th or9th week of administration.Conclusion:(1)High level of oxidative stress based on hyperglycemia will induce "legacy effect" to maintain a long-term damage to the vascular endothelium, which roles as one of the mechanisms of diabetic macroangiopaty.(2)Those oxidative stress indexes as ox-LDL、8-iso-PGF2a and3-NT are closely related to the whole disease.(3)Early enough-course application of SQCP can effectively improve oxidative stress state and further cut off "legacy effect" to finally reduce the likelihood of the occurrence and development of diabetic macroangiopathy with multiple target pionts and paths.
