| Objective:The research based on "the prevention theory in TCM" applys Shenqi compound recipe which could nourish yin,tonify qi and remove blood stasis to the macroangiopathy in spontaneously diabetic KKAy mice,observes the effect on general status,glucose and lipid levels,hs-crp levels and aortic morphology,analyzes the effctive mechanism through miRNA expression.Methods:40 male KKAy mice,7-8 weeks old, random blood glucose≥13.9mmol/L,were divided into 4 groups:the bland group,the ShenQi Compuound Prescription group,atorvastatin group and the diabetic macrovascular disease group.Each group had 10 mice.The model group,ShenQi Compuound Prescription group,and atorvastatin group mice were induced by adding The NO-nitrio-L-arginine methyl ester (L-NAME) with 0.2mg·ml-1·d-1 into the drinking water for 5 weeks. The blank group were fed with normal diet,while others with high fat diet. At the same time, ShenQi Compuound Prescription were administered ShenQi compound recipe with 14.4g/kg-d,and atorvastatin group was administered atorvastatin suspension with 10ml·kg-1·d-1. In the experimental period, general status were observed. After 5 weeks, the mice were sacrificed to detect the fasting blood glucose, lipid levels,hs-crp levels, HE Staining in abdominal aortic for observing aortic morphology, application of Agilent micrornas chip technology platform to test the miRNA expression in aortic tissue,screening differentially expressed miRNAs,And predicting its target genes using TargetScan databases, GO annotations further on the target genes and Pathway of bioinformatics analysis, with a deep understanding of the regulation of miRNAs mechanism and biological function.Results:The general state of ShenQi Compound Recipe group was better than that of model group, with blood glucose being decreased, as well as LDL-c,TG,TC and hs-crp and HDL-c increased.The Differentially expressed miRNAs of ShenQi Compound Recipe group,compared with the model group, are nine in total in which 5 are up-regulated and 4 are down-regulated. After the database predicted target genes,through GO annotation, pathway analysis, the mechanis mainly involved catabolic process.cellular response to stimulus, cell death and the reaction of insulin, signaling pathway,TGF-beta signaling pathway and PPAR signaling pathway.Conclusion:the shenQi Compound Recipe can improve the general state of KKAy mice with diabetic macrovascular disease, improving glucose and lipid metabolism disorders, decreasing hs-CRP, alleviating the vascular endothelial injury, delaying the progress of atherosclerosis. Early application of the shenQi Compound Recipe can prevent and treat the diabetic macrovascular complication effectly by regulate the miRNA to release obesity and insulin resistance, relieve the chronical inflammation of the vessel and inhibit the hypertrophy of the smooth muscle through several genes and pathways. |
