Data Mining Based On Professor Zhou Yabin’s Study In The Treatment Of Cardiac Neurosis And Core Prescription Network Pharmacology Research

Objective:This study uses the Traditional Chinese Medicine Inheritance Assistance Platform 3.0 software with conducting research on Professor Zhou Yabin’s clinical treatment of cardiac neurosis patients,to summarize the medication pattern,and medication characteristics,conducts clinical experience and draw the network pharmacology analysis on the core formulas,explores the mechanism of its treatment of cardiac neurosis,for better guide clinical practice.Method:1.Data mining methodCollecting patient’s cases of cardiac neurosis treated by Professor Zhou Yabin in the outpatient clinic of the First Affiliated Hospital of Heilongjiang University of Traditional Chinese Medicine from June 2014 to July 2023,and all medical cases are screened according to the inclusion and exclusion criteria,921 cases of cardiac neurosis are included and establish a database.Using the Traditional Chinese Medicine Inheritance Assistance Platform 3.0software to extract all the data and analyze the drug association rules.2.Network pharmacologyNetwork pharmacology is used to conduct research based on core prescri-ptions analyzed by complex networks.The main active ingredients and potential targets of the 12 flavors in the core formula were screened by using the BATMAN-TCM database and the TCM Systematic Pharmacological Analysis Platform(TCMSP),and all the relevant genes were retrieved,downloaded and integrated by searching the Genecards database,and then the relevant targets of cardiac neurosis are determined,and then complete the network diagram of the compound-active ingredient-treatment target.Using the String database,the protein-protein interaction network diagram of the compound therapeutic targets was constructed to screen out the core targets.Using David database to analyze the GO functional enrichment analy-sis and KEGG pathway enrichment for the compound therapeutic targets.Research results:1.Data mining results:(1)General condition:There were 921 cases of CN,including 648 female patients,accounting for 70%,273 male patients,accounting for 30%,with a male-to-female ratio of about 1:2.33,and the age of the patients was mainly distributed in the age group of 50-59,with a total of 281 cases,accounting for30.51%.(2)Four diagnostic information:A total of 114 symptoms were involved in 921 cases of CN,and the top 10 symptoms were palpitations,poor sleep,shortness of breath,chest tightness,irritability,fatigue,sweating,dizziness,anorexia,and back pain,accounting for 64.05%.Among them,the rank of top is palpitations,with a total of 712 cases,accounting for 17.74%.According to the statistical analysis results of tongue coating and pulse frequency,tongue red,pale red,purple and dark red were more common,accounting for 75.91%.Moss images were mostly thin white and white moss,accounting for 66.23%.The pulse signs were mostly thin,thin and weak,accounting for 84.58%.(3)Statistical analysis of the medication rules of the group formula’s results:There are 208 of traditional Chinese medicines of total were used in921 cases of CN,with a total drug use frequency of 19196 times,and the top20 drugs with the highest frequency in the overall drug use statistics were cortex et radix polygalae,rhizoma pinelliae,rhizome radix et rhizoma nardost-achyos,ramulus cinnamomi,semen ziziphi spinosae,radix astragali,Codonopsis,Bupleurum chinensis,Paeonia albarum,fossilia ossiamastodi,concha ostreae,baical skullcap root radix scutellariae,radix et rhizoma rhei,blighted wheat fructus tritici levis,rhizoma cyperi,lancelesf lily bulb,fructus schisandrae,Magnetitum,and radix sophorae flavescentis.The four qi are mainly cold and warm;The five flavors are mainly sweet and bitter;The meridians are mostly lung meridians,spleen meridians,heart meridians,and liver meridians;The efficacy of the drug is mainly to replenish deficiency and clear heat.There are a total of 214 association rules,5 groups of core drug combinations,and the commonly used drug pairs are radix bupleuri-ramulus cinnamomi-white paeony root radix paeoniae alba,poria cocos-cocoon,lancel-esf lily bulb-Rehmanniae Radixi,fossilia ossiamastodi-concha ostreae,rhizoma atractylodis macrocephalae-radix aucklandiae,radix astragali-Codo-nopsis;The core drug combination of fossilia ossiamastodi,concha ostreae,rhizoma pinelliae,radix bupleuri,ramulus cinnamomi,white paeony root radix paeoniae alba,rhizome radix et rhizoma nardostachyosi,semen ziziphi spinosae,cortex et radix polygalaei,cocoon,radix astragali,and codonopsis is the core prescription for the treatment of this disease.2.Network pharmacology results:A total of 137 active ingredients(mainly including quercetin,kaempferol,stigmasterol,etc.)acted on 56 targets of cardiac neurosis(mainly involving CHRM1,GABRA1,ESR1,ACHE,etc.)in various databases.GO function enri-chment analysis yielded 292 items of biological process,70 items of molecular function,and 42 items of cell composition,which mainly involved the regulatory activity of signal receptors,RNA polymerase II specific DNA-binding transcription factor binding,membrane microdomain,and hormone response.KEGG signaling pathway enrichment analysis showed that94 pathways were mainly involved in neuroactive ligand-receptor interaction signaling pathway,Yersinia infection-related signaling pathway,lipid and atherosclerosis signaling pathway and AGE-RAGE signaling pathway in diabetic complications.Conclusion:(1)Professor Zhou Yabin believes that the occurrence of cardiac neurosis is closely related to emotional factors,and the pathogenesis of the disease is reflected in the development of the lesion,which is the deficiency of heart,liver,qi and blood,and the standard is mainly phlegm,blood stasis,qi stagnation,qi depression,and fiery heat,which are often mixed with each other.The disease is located in the heart,especially the liver,spleen,lungs,kidneys,and gallbladder are closely related.(2)The core formula is mainly composed of 12 herbs,including fossilia ossiamastodi,concha ostreae,rhizoma pinelliae,radix bupleuri,ramulus cinnamomi,white paeony root radix paeoniae alba,rhizome radix et rhizoma nardostachyosi,semen ziziphi spinosae,cortex et radix polygalaei,cocoon,radix astragali,and codonopsis,which are composed of 12 flavor medicines,through nourishing the heart and liver,soothing the liver and qi,strengthening the spleen and stomach,relieving depression and calming the nerves,and communicating the heart and kidney to reconcile the spleen and stomach Qi machine lifting movement.(3)The core formula contains various active ingredients,which exert its positive therapeutic effect on cardiac neurosis through multi-component,multi-target and multi-pathway.It is speculated that the drugs in the core formula may act on BDNF,IL-6,INS,TNF,IL-1β and other targets through lipid and atherosclerotic signaling pathways,neuroactive ligand-receptor interaction signaling pathways,Yersinia infection-related signaling pathways,and other dopaminergic synapse-related signaling pathways,hormones,inflammation-related signaling pathways,etc.,and sorted out the theoretical basis for the basic research and clinical research application of core prescription for the treatment of cardiac neurosis.
