The Clinical Experience Of Professor Yuan Huishu In Treating Insomnia Disorder Was Summarized Based On Data Mining And Network Pharmacology

Objective:1.The purpose of this study was to summarize the medication rule,dialectical classification,common medication group and prescription characteristics of Professor Yuan Huishu in the treatment of chronic insomnia by TCM inheritance assistance platform.2.In addition,Drug target and Therapeutic pathway of core drugs were verified by network pharmacology,for the development of new drugs,and the theory of medication used by Professor Yuan Huishu in the treatment of chronic insomnia was systematically summarized to provide new guidance for the clinical prevention and treatment of insomnia and new methods for the inheritance of clinical experience of tutors.Methods:1.The effective first diagnosis cases of chronic insomnia treated by Professor Yuan Huishu were screened and recorded into the traditional Chinese medicine inheritance assistance platform(V2.5)after normalization.The common witness type,drug frequency,four Qi and five flavor normalization of drugs and common drug groups were analyzed in the treatment of chronic insomnia treated by the tutor.2.And the core drugs of Professor Yuan Huishu for insomnia were summarized.And pharmacology study through the network on the basis of core drug targets and the disease of chronic insomnia targets,targets,it is concluded that the combination of the communication and constructing PPI network proteins interact with each other,and the use of functions and KEGG pathway enrichment,GO study core drug pathway and chronic insomnia.Results:1.A total of 200 cases were screened and included,among which 154 were female and 46 were male.The majority of patients aged 50-59 were 67,and the top five syndrome types were respectively "Syndrome of liver depression turning fire","syndrome of two deficiency of heart and spleen","syndrome of Yin deficiency and fire boom","syndrome of two deficiency of heart and spleen+liver depression turning fire","liver depression turning fire+Yin deficiency turning fire boom".Frequency of the top five symptom is poor "mei" and "moss thin white","palpitation","pulse string","dream".In the statistics of "four Qi",the frequency of "neutral drugs" was the most,and in the statistics of "five tastes",the frequency of sweet drugs was the most.According to the statistics of the "meridian normalization" of drugs,the top three with the highest frequency were "liver meridian","heart meridian" and"spleen meridian".Drug use frequency statistics,the top ten "tuckahoe"respectively "zizyphus jujube benevolence" "radix bupleuri" "atractylodes""dried tangerine or orange peel" "dang" "polygala" "Acacia skin","oyster","keel","Scutellaria".According to the statistics of the inheritance aid platform "Prescription law",the highest frequency of the two medicines group was "Poria cocos,jujube seed";In the three herbs group,the highest frequency was "Bupleurum,Poria cocos,jujube seed".In the four herbs group,the most frequent ones were "Atractylodes rhizoma,Bupleurum,Poria cocos,jujube seed".In the five-herb medicine group,the most frequent ones were "Tangerine peel,Atractylodes rhizoma,Bupleurum,Poria coporia,jujube seed".Association rules based on drugs,with the highest degree of confidence "angelica root,tuckahoe," "dried tangerine or orange peel Angelica root,tuckahoe,"" dried tangerine or orange peel Angelica radix bupleuri,poria cocos,"" dried tangerine or orange peel atractylodes radix bupleuri zizyphus jujube benevolence and poria cocos".2.The core drugs were poria,jujube kernel,bupleurum,atractylodes,tangerine peel and angelica sinensis.It is based on the core drug.56 core drug active ingredients and 182 drug targets were retrieved from TCMSP database.By searching GeneCards,OMIM,TTD,DISGENET and DRUGBANK databases,1518 targets related to chronic insomnia were obtained.A total of 66 communicative targets of disease targets and drug targets were selected to construct PPI network.The network had 66 nodes,1246 edges,and 10 key targets including CAT,MAPK3,TNF.GO functional enrichment analysis included the response to external stimuli,cell response to organic cyclic compounds,response to nutrient levels,etc.The effects on molecular functions were mainly manifested in G-protein-coupled amine receptor activity,heme binding,and G-protein-coupled neurotransmitter receptor activity.The KEGG pathway enrichment pathways related to insomnia disorder included AGE-RAGE signaling pathway in diabetic complications,chemical carcinogenicity-receptor activation,TNF signaling pathway,etc.Conclusion:1.In the treatment of chronic insomnia,Professor Yuan Huishu focuses on the dialecticality of five zang organs,pays attention to the functions of liver,heart and spleen,and takes into consideration the kidney,lung and three organs as well as three jiao.He holds the three most closely related pathogenesis of chronic insomnia,liver depression turns to fire,heart and spleen deficiency,Yin deficiency turns to fire hyperactivity.2.Network pharmacology studies have shown that in the treatment of chronic insomnia,the core drugs of mentor are active components such as quercetin,stanosterol,kaemptin,naringenin and β-sitosterol,which mainly act on the targets such as CAT,MAPK3,TNF,IL6 and IL1B.Activating AGE-RAGE signaling pathway,chemical carcinogenicity-receptor activation signaling pathway,TNF signaling pathway,cGMP-PKG signaling pathway,etc.in diabetic complications by stimulating membrane rafts,postsynaptic membranes,endocytic vesicles,receptor complexes,and outside of the plasma membrane."It causes responses to external stimuli,cellular responses to organic cyclic compounds,responses to nutrient levels,blood circulation,responses to oxidative stress,etc.,in order to treat chronic insomnia."3.Professor Yuan Huishu’s clinical experience in the treatment of chronic insomnia can be summarized as "nourishing Yin and nourishing the liver","leveling the liver and enhancing Yang","dispersing liver qi","strengthening the spleen and regulating the middle","nourishing Yin and tonifying blood","clearing heat and phlegm","clearing heart and reducing fire","replenishing heart and gall","communicating heart and kidney","promoting blood circulation and removing blood stasis",which is the so-called "ten methods for treating insomnia".
