| Objective:To study the mechanism of acupuncture on skin permeability barrier function,establishing epidermal permeability barrier dysfunction model--guinea pig model of spreading dampness-heat syndrome,blood deficiency and wind dry syndrome and spleen deficiency and dampness accumulation syndrome.Methods:(1)Guinea pigs were divided into blank group,model group,control group and acupuncture group.The control group was given Loratadine tablets by gavage.Acupuncture group was given acupuncture,and basic acupoints are double lung acupoint,double spleen acupoint,double quchi,double Zusanli.(1)guinea pig model of typical disease syndrome of epidermal permeability barrier dysfunction with dampness-heat accumulation of skin syndrome eczema was established.The acupuncture group,basic prescription was added with shuangxuehai,and acupuncture was mainly performed with pingbu and pingxie.The needle was left for 15 min,and suspension pecking moxibustion was performed at shuangfei shu point and shuangzusanli point for 10 min,once a day,for consecutive 7 days.(2)To establish a guinea pig model of typical disease syndrome of epidermal permeability barrier dysfunction,blood deficiency and wind dryness syndrome eczema was established.The acupuncture group,the basic prescription was added to Shuangdishu Point,Shuangxuehai point and Shuangsanyinjiao point,and the acupuncture was mainly performed by the method of reinforcing and eliminating,and the needle was left for 15 min after the puncture,and the treatment lasted for 14 days.(3)The guinea pig model of typical disease syndrome of epidermal permeability barrier dysfunction with spleen deficiency and dampness and eczema was established.In the acupuncture group,basic prescription was added to Yinlingquan point and Shenque point,retaining 15 min,selecte Zusanli,Shengque moxibustion for 10 min,1 times a day,treatment for 14 days.(2)Explore the skin tissue morphology,Water content of stratum corneum(WCSC)and transepidermal Water loss before and after acupuncture intervention.TEWL),serum immunoglobulin E(Ig E),interleukin-4(IL-4),interleukin-17(IL-17)concentrations,AQP3 m RNA and protein expression and distribution.Results:(1)In the guinea pig model of eczema(dampness-heat syndrome): Compared with model group,skin score was decreased significantly(P < 0.01),skin cuticle moisture content(WCSC)was significantly increased(P< 0.01),transepidermal water loss(TEWL)was significantly decreased(P < 0.01),Ig E and IL-17 concentrations were significantly decreased(P <0.01).Il-4 was significantly increased(P < 0.01),skin tissue morphology and structure were improved in sensitized area,and AQP3 expression was increased in sensitized area(P < 0.05).(2)In the guinea pig model of epidermal permeability barrier dysfunction typical eczema(blood deficiency and wind dryness syndrome)experiment,compared with the model control group,The lesion score of skin in acupuncture group was decreased significantly(P < 0.01),and the Water content of the stratum corneum(WCSC)was significantly increased(P < 0.01).Trans-epidermal water loss(TEWL)was significantly decreased(P < 0.01),Ig E and IL-17 were significantly decreased(P < 0.01),and IL-4were significantly increased(P < 0.01).AQP3 expression in skin tissue of sensitized area increased(P < 0.05).(3)In the guinea pig model of epidermal permeability barrier dysfunction,eczema(spleen deficiency and dampness accumulation syndrome)experiment,compared with the model control group,the lesion score of skin in acupuncture group was significantly decreased(P < 0.01),and the skin cuticle moisture content(WCSC)was significantly increased(P < 0.01).Transepidermal water loss(TEWL)was significantly decreased(P < 0.01),Ig E and IL-17 concentrations were significantly decreased(P < 0.01),and IL-4 concentrations were significantly increased(P< 0.01).AQP3 protein expression in skin tissue of sensitized area increased(P <0.05).Conclusion:In guinea pig model with epidermal permeability barrier dysfunction,acupuncture can improve epidermal moisture regulation,improve cytokine expression,play systemic immune function,repair the damaged epidermal permeability barrier function,and further improve the "fur microenvironment" state. |
