The Research On The Prescription Mechanism Of Kidey-qi Bolus And Functions Of Different Proportions To The Mouse Models Of Gentamicin Renal Injury

Kidey-qi bolus,which is composed of eight ounces of dry dihuang,four ounces of shanyao, four ounces of shanzhuyu, three ounces of fuling, three ounces of danpi, one ounce of rougui and one ounce of fuzi ,was created by the medical saint Zhangzhongjing of East Han Dynasty. It’s one of the most famous and effective formulas tonifying kidney and a truly eternal example.Based on the development of history,and the explanation by the previous and contemporary doctors as well as the disease cured by the original formula,The author believe the prescription mechanism of the kidney-qi bolus is used to nourish the kidney-qi rather than nourishing the kidney-yang. The paper makes a research on effects of different proportion on mouse models of gentamicin renal injury to verify the scientitfic the prescription mechanism and what change it will happen if we vary the proportion of kidney-qi bolus.Objective:The experiment uses the method of manufacturing models front the point of view of etiology by gentamicin intraperitoneal injection.We observe the impact of various indicators of the renal injury mouses caused by different proportions. And then try to reveal the mechanism how kidney-qi bolus can tonify kidney-qi and restore the function of kidney controlling water and explore the proportion which is reasonable to treat che renal injury mousse.Methods:The research takes the orthogonal experimental design method and selects the orthogonal table L8(27). The eight kinds of drugs in kidney-qi bolus are devided into four factors and we compose eight prescription according to different doses. Then inject gentamicin into mice’s belly to manufacture models of renal injury and feed each group water decoction of corresponding proportion.Finally observe the impact of indicators of the renal injury mouses.Results:The prescription of four ounces of dry dihuang and four ounces of shanyurou is better than that of eight ounces of dry dihuang and four ounces of shanyurou to reduce the amount of 24-hour urine protein content of mouse models of renal injury.In terms of reducing serum potassium of renal injury mouses,the proportion of eight ounces of dry dihuang is better than that of four ounces.To elevate serum calcium of renal injury mouses, the compatibility of four ounces of dry dihuang and four ounces of shanyurou is better than that of eight ounces of dry dihuang and four ounces of shanyurou ,and the compatibility of one ounce of rougui and one ounce of fuzi is better than that of two ounces of rougui and two ounces of fuzi.On renal tissue nitric oxide synthase, the compatibility of one ounce of rougui, and one ounce of fuzi is better compared with that of two ounces of rougui and two ounces of fuzi to enhance its activity.The compatibility of four ounces of shanyao,and three ounces of fuling can elevate the hemoglobin values in a larger degree than that of three ounces of shanyao and three ounces of fuling.To elevate the total serum protein,the compatibility of eight ounces of dry dihuang, four ounces of shanzhuyu is better than that of four ounces of dry dihuang, four ounces of shanzhuyu, and the compatibility of three ounces of zexie, two ounces of danpi is better than that of three ounces of zexie, three ounces of danpi.Conclusion: The experimental results show that the amount of rougui、fuzi in the original formula is more reasonable .The results confirm that the kidney-qi bolus discussed in the article is used to nourish the kidney-qi rather than nourishing the kidney-yang.It is better to reduce the amount of dry dihuang、danpi in kidney-qi bolus compared with the original formula. When the function of kidney is damaged,it is better to reduce the dose of the medicine,otherwise, it will aggravate the burden of kidney and is not conducive to the treatment of diseases. Shengdihuang is the herb of nourishing yin, but it is sweet ,cold, and zi greasy .In clinic, it is better to reduce the dose of the shengdihuang of the kidney-qi bolus.The reduce of Shanyao is inappropriate,it is better to maintain the amount in the original formula.It may be related to the mild potency of shanyao and the role of nourishing kidney and spleen.The article corrects the use and understanding of kidney-qi bolus which is confused in the clinical application by literature researching. Depending on the development of history, and the explanation by the previous and contemporary doctors as well as the disease cured by the original formula, The author believe the prescription mechanism of kidney qi bolus is restoring the function of kidney controling water,rather than nourshing the kidney-yang. In addition,it confirms that the function of kidney-qi bolus is nourishing kidney-qi,rather than kidney-yang by the animal experiment which is designed in orthogonal aiming at observing the impact of different factors and different levels of kidney-qi on the mouse models of gentamicin renal injury . At the same time,by orthogonal experimental design method we make a research on the impact of functions of different proportions on the mouse models of renal injury and in this way we explain the relationship between dosage and efficacy in traditional Chinese medicine.
