| Objective:Compare and analysis the changes of myocardial morphology,serum TNF-αand cTnI levels in myocardial ischemia reperfusion injury model rats,which accepted exercise preconditioning and electro-acupuncture of"Neiguan" point before and after modeling,And try to discuss the possible mechanism of myocardial protection in the myocardial ischemia reperfusion injury model rats.Method:50 healthy SD rats were randomly divided into 5 groups-control:Sham operation group(S group),Model group(M group),electro-acupuncture group(EA group),Preconditioning group(P group),experimental group(E group).10 rats in each group half male and half female.The rats in S group are raised 21 days and then opened chest with one line surrounded the coronary artery but not ligated,and the rats in the other groups are ligated to establish the myocardial ischemia reperfusion model.M group and EA group are raised 21 days and then established the model.EA group’s then treated with electro-acupuncture on "Neiguan" point(double side).The paremeter of EA is density wave,frequency is 10/50HZ,which is the forelimb quivering as same as the EA,staining 20mins.P group is raised 7 days and then made exercise preconditioning before modeling.The rats in P group swim 15 minutes with the tail 3%weight bearing,and have a rest of 5min,repeat 3 times for 6 days,they will have a rest on the 7th day.Then they have the same excise the second week.Then the modle is established.E group is raised 7days and then accepted exercise preconditioning,continued with electro-acupuncture after modeling.The morphological changes of myocardial tissue were observed with HE staining in each group after the intervention.And the changes of the content of TNF-αand cTnl in the serum of rats were detected by ELISA.Results:1.No morphological changes of myocardial tissue damage in the S group under light microscopy;The morphologic damage of the M group was serious,with lots of inflammatory cells and necrosis of cardiac muscle cells;the cardiac muscle cells of EA group are not regular and the damage of the structure is serious,with some inflammatory cells and necrosis of cardiac muscle cells;the cardiac muscle cells of P group and E group are almost regular and the damage of the structure is not serious,with little inflammatory cells and necrosis of cardiac muscle cells;Compared with M group,the results of morphological observation in the EA/P/E groups were significantly improved.Compared with the EA group,the myocardial morphology of the P group was improved a little.2.The content of TNF-αand cTnI in serum of rats was detected by ELISA,and the results showed that:Compared with the S group,the serum levels of TNF-αand cTnI in all the other groups were increased,and the difference was statistically significant(P<0.01).Compared with the M group,the levels in the EA/P/E groups were all decreased,and the difference was statistically significant(P<0.01).Finally,the serum levels of TNF-αand cTnI in the E group were decreased compared with the P group,and the difference was statistically significant(P<0.01).Conclusion:1.The comparison of the morphological observation results of each group of myocardium under light microscope and the content of TNF-aand cTnI in MIRI model rats serum shows that:Electro-acupuncture at Neiguan point,pre-exercise and exercise preconditioning combined with electro-acupuncture at Neiguan point all can reduce the injury and inflammation of cardiac cells and have certain therapeutic effects on MIRI injury in MIRI model rats.2.Compared with the M group,E group has better morphological changes of myocardial tissue damage under light microscopy and lower content of TNF-aand cTnI in serum of rats.It is proved that:Compared with single exercise preconditioning,electro-acupuncture of "Neiguan" point in rats with myocardial ischemia reperfusion injury after exercise preconditioning has better protective effect... |
