Study On The Intervention Effect And Mechanism Of Self-prescribed Prescription "Maguiye" On Bladder Cancer T24 Cell

Objective: To investigate the inhibitory effect of Magui liquid on the migration and metastasis of human bladder cancer mediated by CC Chemokine ligands 17/CC Chemokine Receptor 4.Methods: 1.The inhibition effect of Magui liquid on bladder cancer T24 cells,as well as its influence on cell morphology and cell cycle,was observed and analyzed by CCK8 method,biological inverted light microscope observation and flow cytometry detection technology.2.Western Blot,QRT-PCR and immunofluorescence were used to detect the expression of CCR4 in human bladder cancer T24 cells and the intervention effect of Magui liquid on CCR4.3.Transwell assay and cell scratch assay were used to detect the effect of Magui liquid on inhibiting the invasion and migration of CCL17 on bladder cancer cells.Results: 1.The Traditional Chinese medicine Magui liquid had significant inhibitory effect on bladder cancer T24 cells.With the increase of drug concentration and the extension of culture time,the viability of T24 cells decreased significantly,and the number of necrotic cells increased in each group,with statistical significance(P<0.05).2.The percentage of T24 cell cycle after treatment with different concentrations of Magui liquid showed statistically significant difference by flow cytometry(P<0.05),and cells were mainly retained in G1 phase after treatment with Magui liquid.3.Chemokine receptor CCR4 was significantly overexpressed in bladder cancer T24 cells;The expression of CCR4 in T24 cells was inhibited by Magui liquid,and the difference was statistically significant(P<0.05).4.Chemokine CCL17 can enhance the invasion and migration of bladder cancer T24 cells and promote the expression of CCR4.Magui liquid can inhibit the invasion and migration of CCL17-mediated BLADDER cancer T24 cells.Conclusion:1.Magui liquid can directly inhibit T24 cells of bladder cancer,and it shows dosetime effect.Magui liquid inhibited T24 cells cell cycle and cell proliferation in G1/S phase.2.Magui liquid solution can inhibit the migration and invasion of HUMAN bladder cancer T24 cells mediated by CCL17-CCR4.The mechanism is to be discussed step in to investigate...
