| Objective:To investigate the protective effect mechanism of Compound Guteng Zhixuan Tablet on hypertensive cardiac hypertrophy and to provide experimental basis for the application of this prescription in the prevention and treatment of hypertensive heart disease.Methods:Of their own spontaneous hypertensive rats(SHR),Wistar rats with normal blood pressure were used as the control group.15 only 8-10weeks of age such as male SHR rats were randomly divided into three groups,respectively is compound uncaria stop being dizzy treatment group(expressed in SHR+FFGT group),l benzene sulfonic acid amlodipine treatment group(expressed in SHR+Levamlodipine group),model group(expressed in SHR group),three groups respectively to compound uncaria daily check anti-dazzle suspension,l benzene sulfonic acid amlodipine suspending liquid and distilled water to fill the stomach.In addition,5 male Wistar rats of the same week age were set as normal blood pressure control group(represented by Wistar group)and given distilled water by gavage daily.In the course of the experiment,the research group regularly measured the weight and blood pressure of rats in each group.Lavage for 8 weeks after treatment,we will kill rats,blood and dissection gather the rat heart left ventricle,left ventricular weight index(LVI)is calculated after weighing,the application of HE staining to observe the myocardial tissue pathological structure,ELISA to detect serum sodium hydrogen permutoid-1(Na~+-H~+exchanger,NHE-1),calcium nerve phosphatase(calcineurin,Ca N),T cell nucleus factor 3(nuclear factor of activated T cell 3,NFAT3)concentrations,Real-time PCR and IHC were used to detect the gene and protein expression levels of NHE-1,Ca N and NFAT3 in myocardial tissue,respectively.Results:(1)Results of blood pressure,body weight and left ventricular mass index:During the experiment period,blood pressure of rats in the Wistar group fluctuated within the normal range;Compared with Wistar group,bloodpressureinSHRgroupwassignificantly increased(P<0.05);Compared with SHR group,the blood pressure in SHR+FFGT group and SHR+Levamlodipine group decreased with the increase of drug intervention time(P<0.05),and tended to be close to normal blood pressure.There was no significant correlation between body weight and blood pressure in each group(P>0.05).After 8 weeks of drug intervention,compared with Wistar group,LVI in SHR group was increased,and the difference was significant(P<0.05).Compared with SHR group,LVI was decreased in SHR+FFGT group and SHR+Levamlodipine group,with significant difference(P<0.05).(2)Compared with the Wistar group,the results of HE staining showed that the left ventricular cardiomyocytes in the SHR group were significantly hypertrophy,nuclear shrinkage and edema,and some of the nuclei of cardiomyocytes disappeared.Compared with SHR group,the morphology and damage of myocardial cells in SHR+FFGT group and SHR+Levamlodipine group were reduced to different degrees.(3)ELISA,real-time PCR and IHC results showed that,compared with Wistar group,the levels of NHE-1,CAN and NFAT3,markers of cardiac hypertrophy in SHR group were up-regulated(P<0.05).Compared with SHR group,the levels of NHE-1,CAN and NFAT3 in SHR+FFGT group were decreased(P<0.05).The levels of CAN and NFAT3 were decreased in SHR+Levamlodipine group(P<0.05),but there was no significant difference in NHE-1 level(P>0.05).Conclusion:(1)Compound Guteng Zhixuan Tablet can significantly reduce the blood pressure of SHR rats and effectively relieve cardiomyopathic hypertrophy,and has the effect of protecting myocardium.(2)The possible mechanism of Compound Goutenzhixuan Tablet in reducing hypertensive cardiomyopathic hypertrophy may be related to the inhibition of NHE-1/CAN/NFAT3 signaling pathway in addition to the effect of lowering blood pressure. |
