Bletilla striata(Thunb.)Reichb.f.Also known as "white peony",it has high medicinal value and high ornamental value.In recent years,the scope of Bletilla striata application has expanded,and the demand for Bletilla striata market has increased greatly.At present,due to artificial over-excavation and destruction of natural habitats,Wild Bletilla striata resources are on the verge of extinction,and the supply of Bletilla striata seedlings is tight,and the means of artificial cultivation that has not yet been effective can widely expand the amount of reproduction.So how tofully exploit the Bletilla striata resources while protecting the existing Bletilla striata resources has become an urgent problem to be solved.This study intends to further optimize the culture conditions of Bletilla striata seed liquids based on plant tissue culture,using Bletilla striata mature hazelnut seeds as raw materials.[Objective]Different levels and combinations of plant growth regulators were used to stimulate Bletilla striata seed,and the culture conditions such as whitening,seed germination and protocorm growth were optimized and screened.The live conditions of the original bulbs were further studied,and the original bulbs were directly broadcasted and seeded live.Compare and explore several key technologies to shorten whitening and growth cycle,reduce production cost and improve seedling quality,and provide scientific basis for the industrial production of Bletilla striata excellent seedlings.[Methods]1.Using the response surface method BBD(Box-Benhnken)test to optimize the concentration of macro elements and growth regulators in liquid medium induced by Bletilla striata seed protocorm.2.Six-factor and five-level orthogonal experimental design was used to screen the best liquid medium for Bletilla striata protocorm growth.3.Screening of matrix and additive species during live broadcast of protocorm by single factor test.4.Observe and compare the Bletilla striata seedling rate and the Bletilla striata seedling quality of the original bulbs and live seed propagation.[Results and Conclusions]1.Optimization of Bletilla striata seed germination liquid culture conditions showed that Bletilla striata seeds were highly adaptable in tissue culture environment,and the medium requirements were not high.From the perspective of the number of protocorms,the best liquid medium for promoting Bletilla striata seed germination was: MS+2.0 mg/L6-BA+0.5mg/LNAA + 0.07 mg/LZT + 30 g/Lsucrose,cultured in the above medium for 45 d,the seed germination rate can reach 93.13%± 0.34%.2.The optimal liquid culture medium for the growth of Bletilla striata protocorm growth is that MS+0.5 mg/L 6-BA+1.0 mg/L NAA+0.02 mg/L ZT+100 g/L banana juice+15 g/L sucrose+15 g/L glucose.3.The optimized culture conditions of the original bulbs showed that the suitable substrate for the original bulbs was 50% humus+0.5 mg/L NAA.The addition of banana mud promoted the growth of protocorm,followed by EM solution and Bletilla striata original slurry.4.The experimental study on the direct seeding and direct seeding of the original bulb showed that compared with the direct seeding of Bletilla striata seed,the original bulb seeding method can shorten the Bletilla striata seedling production cycle,improve the seedling quality and seedling rate,and quickly solve the species.The supply problem of seedlings is suitable for large-scale Bletilla striata seedling production.
