| Objective This study used the public database to analyze the tumor biological data,and combined with experimental verification,to explore the expression of SLC7A11 in renal clear cell carcinoma and its clinical relevance and significance.Methods Download the transcriptome data and clinical data of cc RCC patients from the cancer genome map(TCGA)database and GEO database,and use R software v4.2.1 to collate the data.To analyze the differential expression,prognostic value,enrichment analysis and immune infiltration of SLC7A11 gene in cc RCC,and build a prediction model based on SLC7A11 expression for 1,3,and 5 years.Combined with the collected clinical samples,quantitative real time PCR(q RT-PCR)and immunohistochemistry were performed to verify the results.Results The results of public database analysis showed that the expression of SLC7A11 gene in cc RCC tissue was higher than that in normal tissue(P<0.001),and the expression of SLC7A11 gene was closely related to sex,T stage and distant metastasis(P<0.05).Survival analysis showed that the expression of SLC7A11 gene was significantly correlated with the total survival time of cc RCC patients(P=0.003),and single-factor and multivariate COX regression analysis showed that SLC7A11 gene was an independent risk factor for the prognosis of cc RCC patients(P<0.001).Based on the expression of SLC7A11 and the patient’s age,sex,tumor stage,T stage,lymph node invasion and distant metastasis,the nomograph was constructed.The calibration curve of OS in 1,3,5 years was well fitted with the ideal model,and the C-index index index of the model was 0.778.It was verified by the clinical data of 80 patients,and was well fitted with the prediction model.The enrichment analysis showed that the expression of SLC7A11 in cc RCC was closely related to cell cycle.Immunoassay showed that SLC7A11 was associated with the infiltration of multiple immune cells,indicating that tumor immune microenvironment might play a role in the development of cc RCC.The results of RT-PCR showed that the expression of SLC7A11 gene in cc RCC tissue was higher than that in adjacent tissues.The results of IHC test showed that the expression of SLC7A11 in cc RCC tissue was higher than that in adjacent tissues,and SLC7A11 was expressed in cell membrane and cytoplasm;The expression of SLC7A11 was closely related to age,preoperative symptoms,pathological grade,depth of tumor invasion(T)and clinical stage(P<0.05);The total survival time of the low expression group of SLC7A11 was longer(P=0.003),and the increased expression of SLC7A11 was significantly correlated with the progression-free survival time(P=0.003);Multivariate COX analysis showed that SLC7A11 was an independent prognostic factor for total survival and progression-free survival.Conclusion 1.SLC7A11 is highly expressed in renal clear cell carcinoma and is associated with the clinicopathological characteristics of patients with renal clear cell carcinoma;2.The high expression of SLC7A11 suggests a poor prognosis in patients with renal clear cell carcinoma. |
