Objective:To study the expression of Hsa-miR-5572(miR-5572)in Nasopharyngeal carcinoma(NPC),analyze the relationship between the expression level of miR-5572 and clinicopathological parameters,explore the clinical significance of miR-5572 in nasopharyngeal carcinoma,and explore the potential value of miR-5572 as a new molecular marker of NPC.Methods:1.A total of 82 cases of NPC and 28 cases of nasopharyngeal mucosal chronic inflammation patients who fulfilled the present experimental conditions and were admitted in the People’s Hospital of Guangxi Zhuang Autonomous Region from March 2016 to August 2021 were selected for the study.The NPC group was set as the experimental group,and the chronic inflammation of the nasopharyngeal mucosa were set as the control group.The patients’clinicopathological data were reviewed.2.Quantitative real time polymerase chain reaction(q RT-PCR)was used to detect the expression levels of miR-5572 in FFPE in the experimental group and the control group.The housekeeping gene U6 was used as an endogenous reference.3.Data were subjected to normality test,and non-normal distribution measurement data were expressed as median(interquartile)[M(P25,P75)],and the comparison between two groups was performed using Mann-Whitney U test.Draw receiver operating characteristic curve(ROC)to analyze the diagnostic value of miR-5572 for NPC;Kaplan-Meier and Cox proportional regression risk models were used to describe the survival and prognostic factors of the patients.P<0.05 indicated that the difference was statistically significant.Results:1.qRT-PCR results showed that the expression level of miR-5572 in the experimental group(NPC group)was significantly higher than that in the control group(mucosal chronic inflammation group)(P=0.001);2.The expression level of miR-5572 in NPC tissues was associated with distant metastasis(P<0.05),but not correlated with gender,age,degree of tumor invasion,lymph node metastasis and TNM stage(P>0.05);3.ROC curve analysis showed that miR-5572 predicted the area under ROC curve of NPC diagnosis was 0.716(95%CI:0.611-0.820,P=0.001),the maximum value of Yoden index was 0.384,the corresponding optimal cut-off value was 2.2488,the sensitivity was 63.4%,and the specificity was 75%;4.Kaplan-Meier survival analysis showed that the survival prognosis of the patient with miR-5572 high expression was worse than that of the patients with miR-5572 low expression.COX multivariate regression analysis showed that the high expression of miR-5572 was a risk factor affecting the prognosis of patients with NPC(HR=2.229,P<0.05).Conclusions:1.The expression level of miR-5572 is related to distant metastasis of NPC,which may be an oncogene of NPC.2.miR-5572 may be an independent risk factor affecting the prognosis of NPC patients.3.miR-5572 can serve as a biological marker for the diagnosis and prognosis predication of NPC patients.
