| Objective: The study was designed to investigate the differences in the expression of DNA methyltransferases(DNMTs,including DNMT1/ DNMT3a/ DNMT3b)in three nasopharyngeal carcinoma(NPC)cell lines C666-1,CNE1 and CNE2.Methods: Real-time quantitative polymerase chain reactionPCR(RT-qPCR)was applied to detect the mRNA expression level of the DNMT1,DNMT3 a and DNMT3 b in three nasopharyngeal carcinoma cell lines(C666-1,CNE1 and CNE2)and normal immortal human nasopharyngeal epithelial cell line NP69 as acontrol.The relative quantification of DNMT1,DNMT3 A and DNMT3 B gene expression were measured by 2-△△Ct-Ave analysis method.Results: There were differences significantly in the expression patterns of DNMT mRNAs among C666-1、CNE2、CNE1 and NP69 cell lines.It was observed that the expression level of DNMT1 in turn from high to low was C666-1(32.26±6.15),CNE2(20.26±3.03),CNE1(7.40±0.03)and NP69(1.00±0.10),there were significantly differences in DNMT1 expression level among these cell lines(P<0.01).Moreover,the expression level of DNMT3 a in C666-1 cell line(9.57±0.51)was profoundly higher than that of CNE2(0.81±0.24)、CNE1(1.09±0.14)and NP69(1.00±1.03)(P<0.01),and the comparison of DNMT3 b expression levels were significantly differences among CNE2(3.51±0.06)、C666-1(1.89±0.19)、NP69(1.00±0.11)and CNE1(0.62±0.09)(P<0.01),respectively.Conclusions: It is suggested there are differences in expression patterns of DNMTs in different NPC cell lines which its role and mechanism invoves with epigenetic alterations in the pathogenesis of NPC need to be elucidated. |
