Objective: Download original expression data for bladder cancer through The Cancer Genome Atlas(TCGA).The expression of BGN in bladder cancer was analyzed by statistical and bioinformatics methods,and the expression of BGN in bladder cancer were verified by immunohistochemistry.In order to investigate the expression and clinical significance of BGN in bladder cancer.Method: 1.Download the original data of bladder cancer from the TCGA database,and obtain the basic information and BGN expression of bladder cancer patients after collation.2.The R software program was used to analyze the difference between BGN expression in bladder cancer and normal bladder tissue.3.The BGN expression data obtained from the TCGA database were used to plot the Kaplan-Meier survival curve and analyze the relationship between BGN expression and prognosis in bladder cancer patients.4.Wilcoxon signed-rank test and logistic regression were used to analyze the relationship between BGN expression and clinicopathological features of bladder cancer.5.Univariate and multivariate Cox regression methods were used to analyze the relationship between clinicopathological features and overall survival of bladder cancer patients in TCGA database.6.Gene set enrichment analysis(GESA)was used to predict the regulatory pathway of BGN in bladder cancer.7.To discuss the relationship between BGN expression and immunity in bladder cancer by using Tumor Immune Estimation Resource(TIMER)database and Tumor-immune System Interactions database(TISIDB).8.The Human Protein Atlas(HPA)database and immunohistochemistry experiments were used to verify the difference in BGN expression between bladder cancer and normal bladder tissue in TCGA database.Result: 1.The TCGA database showed that BGN expression was higher in bladder cancer tissue,and there was a difference in expression(P<0.05).2.The expression of BGN increased with the progression of T stage,N stage,clinical stage and pathological grade(P<0.05).3.Logistic regression analysis shows that:Bladder cancer with high BGN expression is more likely to progress to a more advanced stage than bladder cancer with low BGN expression.4.Kaplan-Meier survival analysis showed that the prognosis of bladder cancer with high expression of BGN was worse than that with low expression of BGN(p<0.05).5.Univariate Cox regression analysis showed that there was a significant correlation between bladder cancer with high BGN expression and poor overall survival(HR:1.00;95% confidence interval:1.00-1.00;P<0.05).6.Multivariate Cox regression analysis showed that BGN was independently associated with overall survival(HR:1.00;95% confidence interval:1.00-1.00;P<0.05).7.Gene set enrichment analysis showed that Jak/Stat signaling pathway,Mapk signaling pathway,natural killer cell-mediated cytotoxicity pathway and pathway in cancer were enriched in BGN high expression phenotype.8.The expression of BGN was positively correlated with the infiltration of CD4 T cells,CD8 T cells,neutrophils,macrophages and dendritic cells(P<0.05),and negatively correlated with the infiltration of B lymphocytes(P<0.05).9.There was no significant significant positive expression rate of bladder cancer tissue compared with normal bladder tissue in HPA database(P>0.05).10.The immunohistochemistry results of bladder cancer tissue chip showed that the high expression of BGN protein in tumor tissues was consistent with the expression difference analysis results of BGN m RNA in TCGA database.Conclusion: The high expression of BGN may be a potential prognostic molecular marker of poor survival in bladder cancer.Moreover,the natural killer cell mediated cytotoxicity pathways,pathways in cancer,mapk signaling pathway,jak stat signaling pathway may be the key pathway regulated by BGN in bladder cancer.
