Abstract/Summary:
Objective:Bioinformatics and experimental verification methods are used to find new targeting genes to predict the prognosis of colon cancer,in order to provide new ideas for screening,diagnosis,prognosis evaluation and clinical treatment of colon cancer.Methods:The differential genes in the colon cancer datasets(GSE44076,GSE113513,GSE156355)in GEO database were screened,the differential genes were enriched by GO and KEGG,the protein network interaction map was drawn,and the key genes were screened by Cytoscape software MCODE plug-in.The verification set was used to verify the expression of key genes,COX regression univariate analysis,multivariate analysis and KM survival analysis were performed to analyze the prognosis.ROC evaluated the ability of key genes to predict the diagnosis and prognosis of colon cancer,and drew a nomogram curve model to predict the survival of patients.The wax tissues of 106 patients with colon cancer were collected to make microarrays of cancer tissues and paracancerous tissues.Immunohistochemical experiments were carried out to verify the expression of key genes in colon cancer tissues and paracancerous tissues.The relationship between the expression of key genes and clinical characteristics was analyzed,and the relationship between the expression of key genes and the prognosis of patients was evaluated.Results:A total of 293 differential genes were screened from GEO datasets.The enrichment results showed that the differential genes were mainly involved in Wnt signal pathway,PPAR signal pathway,IL-17 signal pathway and chemokine-related inflammatory signal pathway.The modules containing four genes such as ITM2 C,LRRC19,C1orf115 and TMEM236 were screened by Cytoscape software,and the ITM2 C was selected for study.COX regression analysis showed that ITM2 C was an independent prognostic factor for colon cancer.KM survival analysis showed that the overall survival time of patients with high expression of ITM2 C was better than that of patients with low expression.ROC curve showed that ITM2 C had good diagnostic ability for colon cancer(AUC=0.983).The AUC of 1-,3-and 5-year time-dependent ROC curve were 0.54,0.54,0.65,respectively.The survival prediction model related to ITM2 C is constructed,and the calibration curve shows that the model has better prediction ability.The low expression of ITM2 C in colon cancer was confirmed by immunohistochemistry.Survival analysis confirmed that the survival rate of the group with high expression of ITM2 C was better than that of the group with low expression.Conclusion:The expression of ITM2 C in the adjacent tissues of colon cancer is significantly higher than that in cancer tissues,and the patients with high expression of ITM2 C have a better prognosis.This gene can be used as a potential target for the diagnosis and prognosis of colon cancer.
