| Objectives Bioinformatics and immunohistochemistry were used to detect the expression of ZNF408 in cancer tissues and paracancerous tissues of patients with colon cancer,and to analyze its association with clinicopathological features,so as to find an effective predictor for evaluating the prognosis of patients with colon cancer.Methods The expression and clinicopathological factors of ZNF408 in colon cancer tissues and normal tissues were analyzed by public database.Gene microarray(75 cases of colon cancer tissue microarray and 75 cases of corresponding paracancerous tissue microarray)was used to test the expression level of Zn F408 in paracancerous cel s and cancer cel s by immunohistochemical techniques,and the clinical factors of its occurrence were analyzed and studied.Results 1 In this paper,the relevant experimental data contained in the Timer database,Human Protein Atlas database,UANLCAN database and Oncomine database were used to analyze the relevant data,and result show that the expres of ZNF408 RNA and Protein in patient colon cancer cell was higher than that in normal group(P<0.001).2 UALCAN database analysis showed that there was a significant correlation between cancer stage and the related expression level of ZNF408 m RNA in patients with colon cancer(P<0.001).MRNA expression level was relatively high in patients with colon cancer at the late stage,and the highest expression level was found in the fourth stage.3 Kaplan-Meier plotter surviva l analysis indicated that the high expression of ZNF408 m RNA significantly affected the overall survival of patients with colon cancer(P=0.02).4 String analysis indicated that there was an interaction between ZNF408 and Tspan12,NDP(Norrin),FZD4 and Top3 b,and the correlation scores were 0.824,0.774,0.724 and 0.715,respectively.5 Cbioportal analysis indicated that the mutation rate of ZNF408 in colon cancer patients was 1.9%.6 Timer database analys show that the expres of ZNF408 was negative correlat with the result of B cel s(P<0.05)and CD8+T cel s(P<0.001),and positive correlat with the result of CD4+T cel s(P<0.001).From the above research results,this correlation was of certain statistica l significance.7 The results of immunohistochemistry showed that the positive particles of ZNF408 were expressed in the cytoplasm,the positive expression rate was 100% and the strong positive rate was 74.3% in the cancer tissue,and the positive expression rate was 75.7%and the strong positive rate was 17.6% in the paracancerous tissue,which indicated certain statistical significance(P<0.001).8 The expression of ZNF408 with age(P = 0.04),the infiltration of the depth of the related results(P<0.05)and lymph node metastas(P<0.001),the result of the distant metastasis results(P<0.001),TNM staging results(P<0.001),the degree of differentia(P=0.01)are presented with the corresponding correlation,the correlatio n has statistical significance.Conclusions 1 The expression of ZNF408 in cancer tissue was significant higher than that in normal tissue,and it was an oncopromoter gene of colon cancer.2 High ZNF408 expression affects the overall survival of colon cancer patients.3 The expression of Zn F408 was negatively correlated with B cells and CD8+T cells,while positively correlated with CD4+T cel s.4 The expres of Zn F408 in patient with colon cancer was correlat with the patient age,relevant depth of inva,lymph node metastas,distant metastas,TNM stage and degree of differentia,and the authentic correla was positive correlat.Figure 11;Table 2;Reference 140... |
