| Objective:Transient receptor potential vanilloid acid 1(TRPV1)is widely expressed in human nasal mucosa.With the in-depth study of TRPV1,it is found that it may be widely involved in the occurrence and development of upper airway diseases.However,the function of TRPV1 in nasal epithelial cells and how it affects nasal inflammatory diseases through immunogenic inflammation remains to be further studied.The aim of this study was to determine the existence and expression of epithelial TRPV1 and related epithelial-derived cytokines in chronic rhinosinusitis with nasal polyps(CRSwNP)compared to healthy nasal mucosa,and explore the role of TRPV1 in the pathogenesis of CRSwNP.Methods:Healthy inferior turbinate mucosa(n=10)and nasal polyp tissue(n=20)were collected.Nasal polyps were stained with HE.According to eosinophil count,CRSwNP group was further divided into eosinophilic chronic rhinosinusitis with nasal polyps(Eos CRSwNP(n=10))and non-eosinophilic chronic rhinosinusitis with nasal polyps(non-Eos CRSwNP(n=10)).The patient’s condition was evaluated objectively by Lund-Mackay score before operation.Immunohistochemical staining(IHC)was used to analyze the expression and localization of TRPV1 in normal nasal mucosa and nasal polyps.The expression levels of TRPV1,IL-25,IL-33 and TSLP in each group were quantitatively detected by RT-PCR.The differences between groups and the correlation of indexes were analyzed by Graph Pad9.0software.Results:1.Using IHC staining to localize the expression of TRPV1 in nasal polyps and nasal mucosa tissues of subjects,it was found that TRPV1 was positively expressed in epithelial cells of nasal mucosa and nasal polyps.Compared with the control group and non-Eos CRSwNP group,the expression of TRPV1 in nasal polyps of Eos CRSwNP group was significantly increased,with statistical significance(P<0.05);2.PCR results showed that the expression levels of TRPV1,interleukin-25(IL-25),interleukin-33(IL-33),and thymic stromal lymphopoietin(TSLP)in epithelial cells of Eos CRSwNP group were significantly higher than those of control group and non-Eos CRSwNP group(P<0.05);There was no significant difference in expression between the control group and the non-Eos CRSwNP group(P>0.05);3.The results of Pearson’s correlation rank sum test showed that the quantitative results of TRPV1 in epithelial cells were positively correlated with the Lund-Mackay score in Eos CRSwNP group(R=0.76,P<0.05).Conclusion:1.The expression of TRPV1 in epithelial cells of nasal polyps in Eos CRSwNP patients is increased,and it is correlated with the severity of Eos CRSwNP;2.TRPV1 in nasal polyp epithelium may participate in the pathogenesis of Eos CRSwNP by affecting the epithelial barrier function and the release of epithelial derived inflammatory factors,suggesting that TRPV1 in nasal mucosal epithelial cells have important significance in the pathogenesis of Eos CRSwNP. |
