| Part one: Using bioinformatics analysis to verify the expression of FMNL3 in Hepatocellular carcinoma Background: Primary liver cancer is one of the most common cancers in the world,and it is also an important cause of cancer death.Among them,HCC accounts for 90% of primary liver cancer cases.Studies have found that FMNL3 plays an important role in cell migration and angiogenesis,which is very similar to the biological characteristics of tumor cell invasion and metastasis,and is likely to participate in and promote tumor invasion and metastasis.The purpose of this study was to investigate the expression of FMNL3 in hepatocellular carcinoma and its role in diagnosis and prognosis.Methods: We analyzed the expression level of FMNL3 in hepatocellular carcinoma from Cancer Genome Map(TCGA)database.The R package was used to analyze the RNA sequencing data to determine the differentially expressed genes(DEG)between hepatocellular carcinoma tissues with high and low expression of FMNL3 in TCGA-LIHC database.Spearman correlation analysis was used to determine the relationship between FMNL3 expression and immune cell infiltration.Logistic regression was used to analyze the relationship between the expression of FMNL3 and the clinicopathological features of patients with hepatocellular carcinoma.Kaplan-Meier survival curve,ROC curve,nomogram model and Cox regression analysis were used to evaluate the diagnostic and prognostic value of FMNL3.Results: Compared with the corresponding control group,the expression level of FMNL3 in tumor tissues of patients with liver cancer was significantly increased.DEGS related to FMNL3 is significantly rich in the pathway of Cell metabolism.The expression level of FMNL3 in hepatocellular carcinoma was significantly correlated with immune cell infiltration.COX regression analysis showed that Tumor status(Tumor free and With tumor)is a potentially independent risk factor affecting the ov Immune infiltrationerall survival time(OS)of patients with liver cancer(HR=1.928,p=0.006).ROC curve analysis showed that the expression level of FMNL3 could accurately distinguish between hepatocellular carcinoma and adjacent normal liver tissues.Conclusion: our study shows that FMNL3 is a potential biomarker for the diagnosis and prognosis of liver cancer.Part two: The relationship between the expression of FMNL3 and the prognosis of HCC liver transplantationObjective: To study the expression level of FMNL3 in HCC and to explore the relationship between the expression level of FMNL3 and clinicopathological features and survival prognosis in hepatocellular carcinoma(HCC).To evaluate the important role of FMNL3 in recurrence and metastasis after HCC.Methods: Double Immunohistochemical method was used to detect the expression of FMNL3 in 93 cases of HCC and 56 cases of paracancerous tissues.The detailed clinicopathological features of the patients were collected and the correlation with the expression of FMNL3 was analyzed.The relationship between the expression of FMNL3 and the prognosis of HCC patients was analyzed by Kaplan-Meier survival curve,and the risk factors of survival prognosis after liver transplantation were evaluated by Cox regression analysis.Results: The Double immunohistochemical results showed that the high expression rate of FMNL3 in HCC and paracancerous tissues was 61.3% and 25.0%,respectively,and the difference was statistically significant.The expression level of FMNL3 in HCC was significantly higher than that in paracancerous tissues.The immunohistochemical results showed that the expression level of FMNL3 in HCC was closely related to whether the patient had liver capsule invasion,MVI and vascular tumor thrombus.The expression level of FMNL3 in HCC tissue was not correlated with sex,age,number of tumors,maximum tumor diameter,preoperative AFP level,preoperative treatment,postoperative pathological tumor differentiation and Satellite stove(P>0.05).Spearman correlation analysis results showed that the expression levels of FMNL3 and MVD in liver cancer tissues were positively correlated,and the difference was statistically significant(r=0.389,P<0.01).The median follow-up time of 93 HCC patients undergoing liver transplantation was 44.9(2.7-104.7)months,of which 37 cases recurred and 30 cases died.Kaplan-Meier survival analysis showed that the 1-,3-and 5-year disease-free survival rates of FMNL3 high expression group and low expression group were 66.7%,56.1%,42.8% and 88.9%,83.3%,80.2%,respectively.There was significant difference in1og-rank test(Prun0.0011),while the 1-year,3-year and 5-year overall survival rates of FMNL3 high expression group and low expression group were 80.6%,68.1%,53% and94.4%,86.1%,81.6% respectively.The overall survival rate of the group with high expression of FMNL3 was lower,the survival rate after liver transplantation was lower,and the prognosis was worse than that of the group with low expression of FMNL3.Conclusion: The expression of FMNL3 in hepatocellular carcinoma is significantly higher than that in paracancerous tissues,and the expression level of FMNL3 in hepatocellular carcinoma is closely related to the invasion of liver capsule,MVI and vascular tumor thrombus: the high expression of FMNL3 in hepatocellular carcinoma can reduce the disease-free survival and overall survival after liver transplantation. |
