| Objective:1 Quality evaluation of fresh and dried houttuynia based on e-nose and Gas chromatography-mass spectrometry,we revealed the characteristics and differences of the "fishy smell" of fresh and dried houttuynia,and provided a reference for the establishment of the quality evaluation system of houttuynia based on sensory evaluation technology and the quality control of fresh and dried houttuynia.2 On the basis of the quality study of medicinal materials,the development of the nasal spray of houttuynia was carried out,and the optimal preparation process of the nasal spray of houttuynia was determined through the study of the aromatic water extraction process and the preparation molding process of houttuynia.3 To better control the quality of the preparation,establish the quality standard and fingerprint profile of houttuynia nasal spray.4 To conduct in vitro anti-inflammatory test on the nasal spray of houttuynia to clarify its anti-inflammatory effect.5 To collect effective prescriptions of TCM for nasal diseases,construct a database of TCM medication for nasal diseases based on the TCM heritage computing platform,and conduct medication pattern analysis and prescription mining to provide reference for clinical treatment of nasal diseases and prescription discovery of innovative TCM.Methods:1 The "fishy" odor and volatile components of fresh and dried houttuynia samples were analyzed by electronic nose technique and gas chromatography-mass spectrometry,data modeling and visualization methods such as principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to compare the fresh and dried houttuynia samples.The correlation between the expression of chemical components and the "fishy taste" index was revealed by correlation analysis.2 Formulation process study: The content of methylnonanone,the main active ingredient of houttuynia,was used as an index to determine the best aromatic water extraction process of houttuynia through single-factor test;the selection and dosage of solubilizers,preservatives and PH regulators were investigated to determine the final molding process of houttuynia nasal spray by using the physicochemical or appearance indexes such as clarity and light transmission rate of the preparation.3 General quality checks(characterization,pH value,loading capacity,etc.)were performed on houttuynia nasal spray,and quality standards for houttuynia nasal spray were established,mainly including: thin-layer identification and content determination and fingerprinting.4 At the cellular level,an in vitro inflammation model of RAW264.7(mouse macrophage cells)was induced by LPS(lipopolysaccharide),and the changes in the content of inflammatory mediators NO(nitric oxide)and inflammatory factors IL-6(interleukin-6)and TNF-α(tumor necrosis factor)were measured by enzyme-linked immunosorbent assay to study the anti-inflammatory effects of houttuynia nasal spray.5 Based on the computational platform of TCM inheritance,the classical works such as "Dictionary of Chinese Medicine Formulas","Drug Standard of the Ministry of Health of the People’s Republic of China-Chinese Medicine Formulation" and related modern research literature were used as the basis,and "nasal abyss","nasal congestion" and "nasal drought" were used as the basis."," nasal haggard "," nasal suffocation "," rhinitis "," allergic rhinitis "allergic rhinitis","allergic rhinitis","chronic rhinitis","sinusitis","atrophic rhinitis " etc.as keywords,from which Chinese medicine prescriptions for nasal diseases are screened to build a database of nasal medication.We used the TCM inheritance computing platform to improve the database of TCM treatment of nasal diseases,and to improve the database of nasal medication in terms of "different symptoms","medication method","frequency of medication" and "frequency of prescription".","frequency of prescriptions","core drugs",etc.,to analyze the rules of nasal prescriptions and experience in the use of medicines,"prescription analysis " function,to carry out the analysis of the statistics of the main diseases,the group prescriptions and the characteristics of the use of drugs.Results:1 Electronic nasal and gaseous determination of fresh and dried houttuynia of both,the radar map of 10 sensors of the electronic nose visually depicted the differences in the "fishy odor" characteristics between fresh and dried houttuynia,and the odor differences between fresh and dried houttuynia groups were characterized and differentiated by modeling analysis(PCA,OPLS-DA),screening to determine the 4 main difference sensor indicators W1 W,W2W,W3 C,W5C.GC-MS analysis identified and characterized a total of 90 volatile components from fresh and dried houttuynia,and modeling analysis(PCA,OPLS-DA)overall characterized and distinguished the chemical composition differences between fresh and dried houttuynia groups,and screened 35 major components.The results of correlation analysis showed that nine substances,including d-pinene,(±)-2-pinene,beta-pinene,stigmasterol,(-)-spathulenol,n-nonenal,(Z)-beta-farnesene,lauricene and o-isopropylbenzene,played an important role in the " fishy odor " of houttuynia.In view of the fact that the overall content of volatile components measured in dried houttuynia was greater than that of fresh houttuynia.Since the fresh houttuynia are not easy to be transported and preserved for a long time,the dried houttuynia are better than the fresh ones from the practical production point of view,so the dried houttuynia were selected as the raw materials for the development of the houttuynia nasal spray.2 Determine the preparation process of houttuynia nasal spray as 100 g of houttuynia herb with 25 times the amount of water,distillation and extraction for 5h,collect about 500 ml of the primary distillate,and then redistillation,collect 100 ml of the redistillate,aromatic water add 0.4% polysorbate 80,0.2% potassium sorbate,mix well,and filter through.The houttuynia nasal spray prepared under the optimal preparation and molding process was nearly colorless and clear liquid with aromatic odor,and the pH was 6.7~6.8.3 The quality control methods for the houttuynia nasal spray were established: thin layer identification,determination of methylnonanone content,and fingerprinting quality control methods.15 common peaks were calibrated in the fingerprint spectrum;and the test results showed that the preparation process of different batches of houttuynia nasal spray was stable,and the similarity of fingerprint spectrum of different batches of houttuynia nasal spray was more than 90%.4 The LPS-induced RAW264.7 mouse macrophages were used to successfully establish an in vitro inflammation model for the administration of houttuynia nasal spray.The results of in vitro anti-inflammatory test showed that the concentration of houttuynia nasal spray was less than 50 mg/m L without significant toxicity to cells,and the contents of inflammatory mediator NO,inflammatory factor TNF-α and IL-6 in the model group were much higher than those in the blank group,and the inflammation model was modeled successfully.The secretion of inflammatory mediator NO and the secretion of inflammatory factors TNF-α and IL-6 were significantly decreased with the increase of dose concentration and were inhibited in a dose-dependent manner.The inhibition of secretion of inflammatory mediator inflammatory factor NO was up to 45.23%,and the inhibition of secretion of inflammatory factors TNF-α and IL-6 was up to 69.89% and 82.17%,houttuynia nasal spray significantly inhibited the secretion of NO,IL-6 and TNF-α in inflammatory cells(P < 0.01)and reduced the inflammatory response,and it was correlated with the administered dose showed a positive correlation.5 After screening by searching and combining inclusion and exclusion criteria,a total of588 publications were included,and a total of 217 TCM prescriptions meeting the screening criteria were obtained,constructed a database of Chinese medicine prescriptions for nasal diseases.By analyzing the collected prescriptions in terms of frequency of use,flavor and meridian,and core combinations,it was found that the most used drugs in terms of the four qi were warm(50.09%),cold(28.06%),and flat(16.70%);the most used drugs in terms of the five flavors were pungent(34.15%),sweet(33.30%),and bitter(25.02%);and the most used drugs in terms of the meridian The most used drugs were those entering the lung meridian(27.26%),spleen meridian(16.35%),stomach meridian(13.58%),and liver meridian(12.75%).The top 3 frequency of use were for antiphlogistic(630 times,36%),tonifying deficiency(442 times,26%),and heat clearing drugs(232 times,14%).The top 5 pairs were Magnoliae Flos-Xanthii Fructus(87 times),Magnoliae Flos-Angelicae Dahuricae Radix(81times),Astragali Radix-Atractylodis Macrocephalae Rhizoma(77 times),Magnoliae FlosSaposhnikoviae Radix(74 times),and Xanthii Fructus-Angelicae Dahuricae Radix(69 times)to obtain the core drug network for the treatment of rhinitis.The core drug network for the treatment of rhinitis in Chinese medicine was screened as Magnoliae Flos,Saposhnikoviae Radix,Astragali Radix,Xanthii Fructus,Glycyrrhizae Radix et Rhizoma,,Angelicae Dahuricae Radix,Atractylodis Macrocephalae Rhizoma,Asari Radix Rhizoma.Conclusion:This project based on the electronic nose and gas chromatography,the quality evaluation and difference study of the "fishy smell" and chemical composition of dried and fresh houttuynia were conducted,and the main relevant components of the "fishy smell" were identified,which provided a basis for establishing the quality of houttuynia based on sensory evaluation techniques.Based on the quality evaluation of houttuynia,we determined the preparation process of houttuynia nasal spray,established the quality evaluation method of houttuynia nasal spray,and initially clarified that houttuynia nasal spray can reduce the activity of LPS(lipopolysaccharide)induced cell inflammation through in vitro pharmacodynamic experiments.In addition,a prescription database of TCM nasal medicine was established to analyze the pattern of TCM prescriptions for the treatment of nasal diseases(rhinitis)based on data mining,which provides a reference for mining the experience of effective TCM medication for nasal diseases and further discovery of new TCM prescriptions for nasal medicine. |
