| Dendrobium nobile Lindl.is rare traditional Chinese medical material in our country.In China,the main distribution areas are: Guizhou,Hainan and Guangxi and other tropical and subtropical regions.In the past few decades,D.nobile were used for the treatment of tumors,hyperglycemia,hyperlipidemia and other nervous disease which may be caused by aging,and because of its non-toxic to human body and broad prospect in the clinical treatment,which attracted extensive attention from scholars at home and abroad.Studies found that the sesquiterpenoids and biphenyl compounds in D.nobile have significant biological activity and these compounds have good application prospect,so we want to study on the low polar extraction part of D.nobile.Otherwise,the content of alkaloids component in Dendrobium nobile Lindl is higher than other Dendrobium plants.So far,the research on Dendrobium total alkaloids has mainly focused on the pharmacological effects of the total alkaloids,the research of monormeric compounds isolated from D.nobile are less,so this study wants to extract,separate,and purify the chemical compounds from D.nobile,especially sesquiterpenoids,biphenyl and alkaloids,so that we can enrich the diversity of compounds of D.nobile and provide evidence for their clinical application and drug development.Many modern chromatographic separation technologies such as normal and reduced pressure silica gel column chromatography,Sephadex LH-20,reverse phase silica gel column and HPLC were used to extract and purify petroleum ether and n-butanol extraction of D.nobile.10 compounds were isolated and identified from the petroleum ether and n-butanol extraction of D.nobile,including 3 new compounds and 8 known compounds.There are 3sesquiterpenoids(1,2,3,10),1 fatty acid(6),3 biphenyl(4,8,9)and 3 other compounds(5,7,11).And then the compounds were identified by multiple modern spectroscopic methods such as MS,IR,NMR,UV,X ray crystallization and optical rotation.They are dendroglycoside A(1),dendroglycoside B(2),dendroglycoside C(3),dendronophenol A(4),dibutyl phthalate(5),1-palmitoyl-rac-glycerol(6),3-methoxy benzyl alcohol(7),dendronbibisline D(8),moscatilin(9),dendroterpene C(10),2,2′-bis(4-hydroxyphenyl)propanebis(2,3-epoxypropyl)ether(11).After that,the α-glucosidase inhibitory activity for some of the compounds and fractions were tested.Most of the fractions and two biphenyl compounds(4,8)showed inhibitory activity.This study focued on the compounds of petroleum ether and n-butanol extraction from D.nobile,and provide theory and technical support for the subsequent application research about D.nobile. |
