Study On Probio-X In Adjuvant Treatment Of Elderly Hypertension And Regulation Of Its Intestinal Flora

Objective:To evaluate the adjuvant effect of Probio-X on elderly patients with hypertension and its effect on intestinal flora,and to explore whether there are differences in blood pressure,Alpha diversity,Beta diversity and species composition of intestinal flora before and after treatment,so as to provide new ideas for the treatment of elderly hypertension.Methods:In this study,65 elderly patients with hypertension who were admitted to our hospital from December 2021 to December 2022 were randomly divided into 35 experimental groups and 30 control groups.The enrolled patients received the same antihypertensive treatment.On this basis,the experimental group was given probiotics Probio-X,and the control group was given an equal dose of placebo for 3 months.The changes of blood pressure before and after treatment were compared between the two groups.The fecal samples of all patients before and after 3 months of treatment were collected for 16 Sr DNA high-throughput sequencing analysis,and the changes of intestinal flora before and after treatment were compared between the two groups.Results:1.After treatment,the systolic and diastolic blood pressure levels of the experimental group and the control group were lower than those before the test,and there was a significant difference(P<0.05).2.Comparison of blood pressure levels in the group,the systolic blood pressure and diastolic blood pressure levels in the experimental group were different from those before treatment at 1 month,2 months and 3 months after treatment(P<0.05),and at 2months after treatment and 1 month after treatment,3 months after treatment and 2 months after treatment,3 months after treatment and 1 month after treatment,the difference was statistically significant(P<0.05).There was no significant difference in diastolic blood pressure between 3 months and 2 months after treatment in the control group(P>0.05).3.Comparing the blood pressure drop before and after treatment in the two groups of patients,compared with the control group,the systolic blood pressure in the experimental group decreased significantly after 3 months of treatment and before treatment(d0-3)(P=0.043);compared with the control group,the decrease of diastolic blood pressure(d2-3)in the experimental group was statistically significant(P=0.032).4.Alpha diversity analysis:There was no significant difference in Alpha index between the two groups before treatment(P>0.05).At 3 months after treatment,there were significant differences in Ace and PD_whole_tree indexes between the two groups(P=0.028;P=0.001);the PD_whole_tree index of the experimental group before and after treatment was statistically significant(P=0.030).There was no significant difference in the control group(P>0.05).There was no significant difference in Shannon and Simpson index between groups(P>0.05).5.Anosim analyzed the Beta diversity between different groups of samples.The difference between the experimental group and the control group after 3 months of treatment and the experimental group before and after 3 months of treatment was statistically significant(P<0.01).6.Analysis of significant differences between groups:Using Lef Se analysis,at month 0,the species with significant differences in intestinal flora abundance between the experimental group and the control group were Fusobacteriota,Firmicutes,Fusobacteria,Fusobacteriales,Fusobacteriaceae,and unclassified Cetobacterium.Among them,Fusobacteria,Fusobacteria,Fusobacteria,Fusobacteria,and unclassified cetobacter are the flora that play an important role in the experimental group;after three months of different interventions,the species with significant differences between the two groups were Bacteroidaceae,Bacilli,and Bacteroides.The microbial groups that played an important role in the experimental group were Bacteroidaceae and Bacteroides.After treatment,Fusobacteria,Fusobacteria,Proteobacteria,and unclassified cetaceans in the probiotic group were significantly lower than those before treatment(P<0.01).Conclusion:1.Probiotic Probio-X has a better therapeutic effect on systolic blood pressure after 3 months of adjuvant treatment compared with antihypertensive drugs alone in elderly hypertensive patients.2.For the decrease of diastolic blood pressure after 3 months of treatment and 2 months of treatment,Probio-X combined with antihypertensive drugs has a better therapeutic effect on elderly patients with hypertension than antihypertensive drugs alone.3.Probiotic Probio-X adjuvant therapy can improve the structure of intestinal flora in elderly patients with hypertension,increase Bacteroidaceae and Bacteroides,and decrease Fusobacteria,Fusobacteria,Proteobacteria and unclassified Cetobacterium.
