| Objective:In this study,fecal and blood samples from patients with ACS and healthy people were tested and analyzed to evaluate the difference of intestinal flora between patients with ACS and healthy people,and to investigate whether ACS is related to intestinal flora,it provides important evidence and possible intervention target for the prevention and treatment of ACS by regulating intestinal flora.Methods:Twenty-eight patients with ACS and 30 healthy people hospitalized in the general medicine ward and emergency internal medicine ward of the Affiliated Hospital of the inner Mongolia medical university from December 2021 to February 2023 were selected as the research objects.Clinical data of ACS patients and healthy people were collected for comparative analysis.Stool and blood samples of 58 subjects were collected,and intestinal flora of stool of the subjects was analyzed by 16SrDNA high-throughput sequencing to compare the difference of intestinal flora between ACS patients and healthy people and its correlation with the incidence of ACS.Results:1.In this study,there were no significant differences between the ACS group and the control group in age,sex,nationality,BMI,hemoglobin(HGB),platelet(PLT),blood glucose(FBG),liver function(ALT),kidney function(Cr),blood lipid(LDL-C)and coagulation function(PT-sec)(P>0.05);The level of cTnT-hs in ACS group was higher than that in control group(P<0.05)2.In this study,the intestinal flora of ACS patients and healthy people were sequenced by the 16SrDNA v3+v4 highly variable region,then OTUs clustering and species annotation were conducted.The Alpha diversity and Beta diversity of annotated species were analyzed,and it was found that there were differences in intestinal flora between the two groups,among which species diversity was not significant(P>0.05),but species abundance was significantly different(P<0.05).Further analysis of the difference between the two groups showed that:(1)At the phylum level,the relative abundances of Proteobacteria,Verrucomicrobiota,Patescibacteria,Gemmatimonadota and Cyanobacteria were significantly different between the two groups(P<0.05);Compared to the control group,The abundances of Proteobacteria,Verrucomicrobiota,Patescibacteria,Gemmatimonadota and Cyanobacteria in ACS group were decreased.(2)At the class level,the relative abundances of Gammaproteobacteria,Negativicutes,Alphaproteobacteria,Verrucomicrobiae,Saccharimonadia were significantly different between the two groups(P<0.05);Compared with the control group,the abundance of Negativicutes in the ACS group was increased,while the abundances of Gammaproteobacteria,Alphaproteobacteria,Verrucomicrobiae and Saccharimonadia in the ACS group were decreased.(3)At the order level,the relative abundances of Lachnospirales,Enterobacterales,Veillonellales,Christensenellales,Burkholderiales,Clostridia and Clostridiales were significantly different between the two groups(P<0.05);Compared with the control group,the abundance of Lachnospirales,Burkholderiales and Veillonellales in the ACS group were increased,while the abundance of Enterobacterales,Christensenellales,Clostridia and Clostridiales in the ACS group were decreased.(4)At the family level,the relative abundance of Lachnospiraceae,Enterobacteriaceae,Christensenellaceae,Clostridiaceae and UnclassifiedClostridiaUCG014 were significantly different between the two groups(P<0.05),the abundance of Lachnospiraceae was increased in ACS group,while the abundance of Enterobacteriaceae,Christensenellaceae,Clostridiaceae and UnclassifiedClostridiaUCG014 were decreased in ACS group.(5)At the genus level,the relative abundances of subdoligranum,Escherichia,Agathobacter,Christensenellaceae,Blautia,Roseburia were different between the two groups(P<0.05).Compared with the control group,the abundances of Agathobacter and Roseburia were increased in ACS group,while the abundance of Subdoligranulum,EscherichiaShigella,Blautia and Christensenellaceae were decreased in ACS group.(6)At the species level,the relative abundance of unclassified Subdoligranulum and unclassifiedEscherichia,unclassified ChristensenellaceaeR7group,unclassifiedBlautia and unclassified Lachnoclostridium were significantly different between the two groups(P<0.05).Compared with the control group,the abundance of unclassifiedLachnoclostridium in ACS group was increased,while the abundance of unclassifiedSubdoligranulum,unclassifiedEscherichiaShigella,unclassifiedChristensenellaceaeR7group and unclassified Blautia in ACS group were decreased.(7)Patescibacteria and Saccharimonadia are associated with cTnT-hs.Conclusion:(1)There was significant difference in species richness between ACS patients and healthy people(P<0.05).(2)Patescibacteria and Saccharimonadiaare are related to the level of cTnT-hs,which may be related to the pathogenesis of ACS.(3)Regulating the gut microbiota of ACS patients according to the functional characteristics of different bacteria may play a positive role in the progression and prognosis of ACS patients. |
