Abstract/Summary:
Spinal cord injury(SCI)often occurs from a series of high-energy mechanical injuries such as car accidents,falls from height,and violent cases,and the injury is often fatal.The most common fatal site was the neck in 61%,followed by the waist in23%,and the chest SCI in 14%.Patients with severe injuries are often difficult to obtain timely and effective treatment.Even if they survive severe injuries,severe complications will significantly reduce the quality of life of patients,and inevitably make the patients,their families and society bear a large burden.In recent years,in order to treat SCI,many research teams have carried out the experimental direction of combining bioengineering materials with stem cells.Nanomaterials have shown many significant advantages in the treatment of SCI.For example,different from traditional intravenous drugs,nanomaterials can avoid the obstruction of blood spinal cord barrier for drugs,directly attach to the lesion area,and play the role of bridging and filling the lesion cavity.At the same time,nanomaterials can act as a very excellent functionalized ligand to provide a vehicle for growth factors,enzymes,anti-inflammatory factors and other substances.Due to its good biocompatibility,it can be stably released after reaching the injury site without affecting the functional performance of the ligand.Ectomesenchymal stem cells-conditional culture medium equipped with Poly(lactid-co-glycolide)acid(PLGA)was investigated medium,EMSCs-CM)in the treatment of SCI.We selected a specific stem cell population that is easily harvested from the mammalian nasal cavity and,like other types of MSCS,has potent immunomodulatory functions.In this experiment,EMSCs were isolated from the nasal cavity of mice and cultured,and the EMSCs supernatant was collected as EMSCs-CM.EMSCs-CM was purified by centrifugation and made into lyophilized powder after dialysis membrane.The lyophilized powder of EMSCs-CM was incorporated into PLGA particles by water-oil-in-solid emulsion method to prepare PLGA particles based on EMSCs-CM.Then implanted into the injured site of healthy Sprague dawley(SD)rat animal model,According to the behavioral score of spinal cord injury(Basso-Beattie-Bresnahan,BBB),histological observation of spinal cord injury(hematoxylin-eosin staining,immunohistochemistry),and protein expression in spinal cord tissue,the recovery of lower limb movement in SCI rats was evaluated.Based on this study,it can be considered that PLGA particles based on EMSCs-CM are very attractive candidates for the treatment of SCI by promoting nerve regeneration,inhibiting inflammatory response,and improving motor function.
