| Objective: To investigate the expression level and diagnostic value of miR497-5p in patients with Alzheimer’s disease(AD),analyze the possible correlation between miR497-5p and fibroblast growth-in-2(FGF-2),and analyze the function and signaling pathway of miR497-5p target genes involved in the occurrence and development of AD by bioinformatics technology.Methods:(1)The serum samples and clinical data of 50 AD patients and 37 control patients were collected.The expression level of miR-497-5p in the serum samples of the two groups was detected by fluorescence quantitative PCR,the FGF-2 level was detected by ELISA kit,and the cognitive function of AD patients was assessed by MMSE scale.Online target gene prediction website predicted the relationship between miR-497-5p and FGF-2,analyzed the correlation between serum miR-497-5p and MMSE score and FGF-2 level,and drew ROC curve to evaluate the diagnostic value of serum miR-497-5p in AD.(2)Target genes of miR-497-5p were found using Target Scan and miRWalk target gene prediction website,and their intersection was selected.Then,verified miR-497-5p target genes were screened from miRTar Base database,and the selected target genes were combined by Venn diagram.The GO and KEGG pathway enrichment analysis of miR-497-5p target gene function was performed by using the DAVID database.Results:(1)The expression level of serum miR-497-5p in AD group was significantly higher than that in control group,and the difference was statistically significant(P<0.01),there was a targeting relationship between miR-497-5p and FGF-2,and serum miR-497-5p was negatively correlated with MMSE score and FGF2 level(r =-0.724,r =-0.748,P<0.01);ROC curve showed that the areas under the curve of serum miR-497-5p for the diagnosis of AD were 0.785(95%CI:0.696 ~ 0.829,P<0.05),sensitivity was 82.7%,specificity was 78.6%.(2)A total of 195 candidate target genes were obtained by three target gene prediction sites.GO and KEGG enrichment showed that miR-497-5p target genes may be involved in chondrocyte differentiation,BMP signaling pathway,cell division,neuronal differentiation and other biological processes,mainly located in the nucleus and cell membrane,cytoplasm and neuronal cell body.It has molecular functions such as transforming growth factor receptor binding,REDOX enzyme activity,phosphatase binding and ATP binding,and is involved in the regulation of cancer pathway,cell aging,m TOR signaling pathway,Alzheimer’s disease,PI3K-Akt signaling pathway and other pathways.Conclusion: Serum miR-497-5p is highly expressed in AD patients,and its target genes can be involved in the occurrence and development of AD by affecting protein kinase activity and cancer signaling pathways,suggesting that miR-497-5p may be a potential biomarker for the auxiliary diagnosis of AD. |
